CLA could induce cell apoptosis and reduce fat deposition in animals,but the specific mechanism was unknown.The paper discussed the effects of different dietary CLA levels on serum lipid metabolic indicators and adipose tissue cell apoptosis in sheep by animal nutrition technology and molecular biology methods and its function.18 sheep were divided into 3 groups randomly,with 6 sheep in each group.They were the control group,the 1.5% CLA group and the 3.5% CLA group.The trial period was 75 days,the pre-feeding period was 15 days and the normal feeding period was 60 days.The test is divided into the following contents:Experiment 1: The effect of different dietary CLA levels on the biochemical indicators related to lipid metabolism in sheep serum.The results showed that there was no significant difference in the content of TC,TG and HDL among the three groups(P>0.05).Compared with the control group,the LDL content in the 3.5% CLA group was significantly decreased(P<0.05).Experiment 2: The effect of different dietary CLA levels on the apoptosis of sheep adipose tissue cells.1)The effect of different dietary CLA levels on the morphological and biochemical characteristics of sheep adipose tissue cells.The results showed that there was obvious yellow-green fluorescence in both test groups by TUNEL staining,and with the increase of CLA content,the characteristics of fat tissue cell apoptosis became more obvious.There were obvious DNA ladder-like bands in 3.5% CLA group by agarose gel electrophoresis.The results showed that the apoptosis of adipose cells occurred,while the other two groups were not obvious results.2)The effect of different dietary CLA levels on the expression of apoptosis-related enzymes in sheep adipose tissue cells.The results showed that there were no significant differences in the enzyme activities of Caspase-3,Caspase-8 and Caspase-9 among the three groups(P>0.05).3)The effect of different dietary CLA levels on the expression of related genes in sheep adipose tissue cells.The results showed that,compared with the control group,the m RNA expression of Caspase-3 and Cyt-C in the 1.5% CLA group was significantly reduced(P<0.05),and the m RNA expression of Bcl-2 was significantly increased(P<0.05),and there was no significant difference in Bax m RNA expression(P>0.05).Compared with the control group and the 1.5% CLA group,the m RNA expression of Bax,Caspase-3 and Cyt-C in the 3.5% CLA group was extremely significantly increased(P<0.01),and the m RNA expression of Bcl-2 was extremely significantly reduced(P<0.01).4)The effect of different dietary CLA levels on the expression of related proteins in sheep adipose tissue cells.The results showed that,compared with the control group,the protein expression of Caspase-3 and Cyt-C in the 1.5% CLA group was significantly reduced(P<0.05),and the protein expression of Bax and Bcl-2 has no significant difference(P>0.05).Compared with the control group and the 1.5% CLA group,the protein expression of Bax,Caspase-3 and Cyt-C in the 3.5% CLA group was extremely significantly increased(P<0.01),and the protein expression of Bcl-2 was extremely significantly reduced(P<0.01).In summary,under the conditions of this experiment,the LDL content was significantly decreased in the serum of sheep in 3.5% CLA group,and increased the apoptosis of the sheep’s adipose tissue cells.While the effect of 1.5% CLA on them needs further research.
