In this study,the LC-MS / MS method was first used to analyze the egg proteomics of T.parvum,and based on the detected data and Uniprot data,1,116 specific peptides and 278 proteins were identified,including 94 highly reliable proteins.According to the nature and function,these highly trusted proteins are classified into 6 categories including Transporters,Enzymes,Peptidase Inhibitor,Putative Secreted Proteins,Putative Housekeeping Proteins,and Uncharacterized protein.Among them,housekeeping proteins are the most,with 25 kinds,followed by secreted proteins,a total of 18 Species.Some yolk proteins(such as Serpins,Vitellogenin)have obvious familiality.Using the proteomic results of T.minichis as a clue and designing primers with known sequences,RACE method cloned the coding genes(m RNA)of Heme-binding asparlic proteinase(THAP),Ferritin,Vitellin-degrading cysteine endopeptidase(VTDCE)and Crystallin),The results show that: THAP gene has a total length of 1381 bp,an ORF box of1010 bp,and encodes a peptide segment of 372 aa;Ferritin gene has a full length of 961 bp,ORF box of 566 bp,and encodes a peptide segment of 190 aa;VTDCE gene has a total length of 961 bp,and an ORF box of 340 bp,encodes a peptide segment 115 aa;Crystallin gene has a total length of 841 bp,an ORF frame of 451 bp,and encodes the peptide 152 aa.The relative expression levels of THAP,Ferritin,VTDCE and Crystallin genes in T.microphyllum were analyzed by RT-PCR.The results showed that the expression of THAP was the highest,followed by VTDCE and Ferritin.the lowest is Crystallin.
