Objective: Genetically modified animals are the development direction of laboratory animals in the future,the development of genome-editing technique gives a great push to the development of genetically modified animals.Pigs have many physiological and biochemical characteristics similar to those of humans,genetically modified pigs are ideal laboratory animal model and ideal donor animals of xenotransplantation.It’s an important task that assessment for biosafety of genetically modified pig in the process of the pigs’ breeding and using.the insulin gene humanized modification boar.Human modification of porcine insulin gene can provide a more ideal donor source for clinical islet xenotransplantation,solve the short-term problem of islet homeotransplantation donor,and provide sufficient human insulin for the treatment of diabetes.The object of this dissertation is one insulin gene humanized modification boar.Be aimed to explore the argument of semen,genetic endowment condition,growth and development condition of their offspring,and comparing them with general pigs,analyzing the difference between the two kinds of pigs,for preliminarily evaluating the safety of the modification pig.To collect the basic data and demonstrating the feasibility for breeding insulin gene humanized modification pig strain.Methods: Selecting 3 general boars which are even-aged and similar physical condition with the modification boar for some time.Sampling and testing repeatedly about evaluating the semen of the 4 boars,in the meantime,choosing some sows which have excellent reproductive performance,and recording mating and parturition information.weigh the birth weight and weight at age in 7 day,14 day,21 day,28day,45 day and 2 mouth of the offsprings.All the date are testing the significance by SPSS.Testing the target gene by ear tissue of the offspring in the sequencing company,and analyzing the results.Results: It’s no significant difference in the semen between the genetically modification pig and general pig(P>0.05).It’s no significant in the growth and development between the two kinds of pigs’ offspring at the two months after birth(P<0.05).The target gene was found in the all offsprings of genetically modification pig.Conclusions: The semen of insulin gene humanized modification boar are “same as” the general boar.The growth and development of the offspring of the insulin gene humanized modification boar are “same as” the offspring of general boar.The humanization gene of the modification boar can normally pass on to offsprings,it has no obvious influence on the growth and life of the offsprings.The insulin gene humanized modification boar has high security,no obvious abnormality has been found at present.The modified boar can be used as the basis for breeding the insulin gene humanized modification pig strain.
