China is the origin of tea and the birthplace of tea culture.In recent years,with the improvement of China’s economy and the enhancement of science and technology,more and more people have higher requirements on the quality and yield of tea,and the promotion of the breeding of clones of tea clones has received more and more attention.Cutting is the most commonly used propagation method for tea plants,it can effectively retain the excellent traits of the female parent,the variety has high purity,and the method is simple and easy to operate.However,due to the long rooting time,the cutting and seedling raising cycle is longer,which affects the promotion efficiency of the excellent tea varieties.In order to accelerate the rooting speed of tea cuttings,shorten the tea planting cycle and reduce the cost of seedlings,this study optimized the cutting conditions of tea plants.First,the influencing factors of adventitious formation of tea cuttings were analyzed,and the influence of factors such as matrix composition,environmental temperature,plant growth agent etc.on the rooting efficiency of cuttings was investigated,and the parameters of each factor affecting the rooting of tea cuttings were determined by a single factor test range.On this basis,the response surface analysis method was used to optimize the analysis of the interaction between various factors,and a mathematical model for rooting efficiency of tea tree cuttings was constructed.According to the model,the optimal rooting culture conditions for tea cuttings were obtained.The followed research results:1.Clarified the influence of substrate on the rooting efficiency of tea cuttings.Studies have shown that different substrates had significant effects on the rooting rate of tea tree cuttings.River sand was the most effective substrate to promote the rooting survival of tea tree cuttings.The rooting rate after 8 weeks of rooting culture was 48.89±1.92%,followed by loess,vermiculite and nutrition earth.The mixing of sand and soil can further improve the rooting efficiency,and the different ratio of sand to soil also had a significant effect on rooting of tea tree cuttings.When the sand content was 70%,the rooting rate of tea tree cuttings within 8 weeks was 52.22±2.55%.2.Clarified the influence of temperature on the rooting efficiency of tea cuttings.Studies have shown that temperature had a relatively large influence on the rooting efficiency of tea cuttings.With the increased of temperature,the rooting rate of tea cuttings gradually increases.When the temperature was 24 degree Celsius,the rooting effect was most significant,when the temperature continued to rise to 30 degree Celsius,the high temperature reduced the physiological functions of tea plants,and the rooting rate of cuttings no longer increased.3.Clarified the effect of plant growth agent on the rooting efficiency of tea cuttings.Studies have shown that rooting agents can significantly increase the rooting rate of tea tree cuttings,and the rooting rates of different types and concentrations of tea tree cuttings varied greatly.Among them,50mg/L SGJ3 rooting agent had the best treatment effect,and the rooting rate of tea tree cuttings can be reached 73.33±6.01% for 3 weeks.4.Clarified the effect of rooting agent treatment time on the rooting efficiency of tea cuttings.Studies have shown that the treatment time of the rooting agent had a significant effect on the rooting time of tea tree cuttings.The rooting rate of tea tree cuttings was the highest at 9 minutes,and the rooting rate of tea cuttings can reached 85.56±3.85% within 3weeks.5.Determined the best rooting culture conditions for tea tree cuttings.Response surface analysis showed that the order of the influence of various factors on the rooting rate of tea cuttings was: soaking time> sand content> rooting agent concentration> temperature;the mathematical model of the rooting rate of tea cuttings through various factors was:R1=88.59-0.19A+0.0888-0.19C+0.51D+0.24A×B-0.085A×C-0.040A×D-0.19B×C-0.85B×D-0.19C×D-0.92A2-0.83B2-1.34C2-1.65D2.According to the mathematical model,the optimal cutting conditions were determined as follows: sand content was 69.45%,temperature was 23.89 degree Celsius,reagent concentration was 54.33 mg / L,soaking time was 8.41 min.Under this condition,the theoretical rooting rate of tea tree cuttings in 3 weeks was 88.66%.Three verification experiments showed that the average actual 3-week rooting rate was 88.4%,which was basically consistent with the predicted value.
