| Medicago falcata L.,a perennial herb in the Medicago,is often used as a gene pool for Medicago breeding.Based on the results of transcriptome sequencing of Medicago falcata L.,a c DNA fragment of AP2/ERF family gene differentially expressed under drought and salt stress,was analytically cloned and named MfERF028(672bp)(Gene Bank No.MW766361).The expression patterns of this gene in roots,stems and leaves under freezing,salt,drought and ABA stresses were analyzed using Q-PCR.The results showed that the expression of MfERF028 in roots,stems and leaves changed after different times of abiotic stresses and was particularly intense in response to-8°C and ABA stresses.MfERF028 was identified as localized in the nucleus using laser scanning confocal microscopy.The promoter of the MfERF028,2385 bp long,was obtained by Genome Walking.The promoter has elements that respond to Me JA,abscisic acid,light,growth hormone,low temperature and a drought-induced MYB binding site,and transiently expresses p BI121-p MfERF028::GUS in tobacco,demonstrating that the promoter has initiation activity.The function of the MfERF028 in regulating plant resistance to abiotic stress was investigated using the hair root transformation method,and the results showed that the root length growth of transgenic plants overexpressing the MfERF028 was significantly higher than the control group after cold stress,and the expression of the Mt CAS15 A was reduced after inhibition of the MfERF028 expression,and the study initially indicated that the MfERF028 has a positive role in plant.Under ABA stress,the results show that MfERF028 gene plays an inhibitory role in the process of Mt RD2 gene expression.In addition,three Medicago sativa L.plants with the MfERF028 were obtained using Agrobacterium-mediated genetic transformation method,which laid the foundation for further identification of gene function. |
