| To understand the starch content and composition of the faba bean germplasm resources and the related coding gene information.Taking 319 faba bean germplasm resources at home and afaba as the research materials.The total starch content,the amylose content and the amylopectin content in 2019 and 2020 were determined and the diversity was analyzed;Qing Can14 material as the research object to expand the faba bean Waxy gene and conduct bioinformatics analysis;Screening 85 faba bean germplasm resources with large difference in amyarch content.Expand the Waxy gene protein coding area with DNA as the template,analyze its nucleotide sequence diversity and phylogenetic tree construction;Finally,filter the SNP sites and perform a haplotype analysis,The polymorphism site was associated with the two-year starch content of 85 germplasm resources.The specific results are as follows:1: The total starch content of 319 materials of 19.208 % ~ 57.472 %,amylose content of 4.148 % ~ 17.181 %,amylopectin content of 15.008 % ~ 45.08%,066%,variation coefficient of starch content was high and diversity index greater than 2.Through clustering analysis of 319 materials were based on starch content,the results showed that all materials were divided into three categories,including the second class content of 237 materials.2: Waxy gene CDS length 1083 bp,encodes 360 amino acids;Waxy protein contains two conserved domains: Glyco_transf_5(PF08323)starch synthase catalytic structure and Glycos_transf_1(PF00534)glycosyltransferase Group1,which is a stable and soluble hydrophilic protein with no transmembrane structure acting on the cytoplasm.3: 85 materials Waxy gene nucleotide Diversity Pi: 0.00733>0.005,construction phylogenetic tree,the results showed that 85 materials were divided into three categories,no obvious regional division,indicating that the Waxy gene is more conservative.4: Waxy protein coding sequence.Through screening,52 core variation sites were obtained.Group structure analysis based on the screening SNP site showed that 85 materials were divided into three categories,with no regional classification.Haplotype analysis revealed that a total of 30 haplotypes were detected in 85 coding sequences.5: The core SNP sites and starch content were analyzed with 4 SNP sites significantly associated with straight amych and 1 SNP site significantly associated with amyarch content.The first 4 sites are in sequence noncoding regions and are not involved in editing the Waxy protein amino acid sequence is a nonfunctional site.The last site in the CDS sequence,located at 493 bp,When thymine T changes to cytosine C,its editing amino acids did not change,still edit K(methionine)is a synonymous mutation. |
