| Spermatogenesis is the key to determine the quality of semen,which is closely related to the fertilization rate in goose.Spermatogenesis is influenced by many factors and differs remarkably between different goose breeds.Although both Sichuan white goose and Lande goose belong to the medium-sized goose species,there observed big differences in their fertilization rate,which may be related to differences in their spermatogenesis.Therefore,by comparing and analyzing the differences in the testicular morphological structure and mRNA-lncRNA expression patterns between male Sichuan White Geese and Lande Geese,we aimed to preliminary explore the molecular mechanism underlying their different spermatogenic ability The main results of this study were listed as follows:1.The organ index of testis and the long diameter of seminiferous tubules of Lande geese were significantly higher than those of Sichuan White Geese(P < 0.05),and the length of external genitalia of Lande geese was significantly shorter than that of Sichuan White Geese(P < 0.01).2.Compared with Sichuan white geese,there were 462 differentially expressed mRNAs(173 down-regulated and 289 up-regulated)and 329 differentially expressed lncRNAs(280 up-regulated,49 down-regulated)observed in Landes geese.Among all these differentially expressed mRNAs,10 genes with FPKM greater than 10 were involved in spermatogenesis,and among them,in addition to the SEPP1 gene,the remaining were significantly up-regulated in Lande geese.3.These differentially expressed mRNAs between Sichuan white geese and Lande geese were significantly enriched in 130 GO terms and 13 KEGG pathways,of which the most significant pathway was the phosphatidylinositol signaling which isan important signaling pathway during spermatogenesis,and all of these differentially expressed mRNAs enriched in this pathway and its related pathway—inositol phosphate metabolism pathway was significantly up-regulated in Lande geese.4.These 329 differentially expressed lncRNAs predicted 1018 cis-targeting genes and 9221 trans-target genes.The 1018 cis target genes were significantly enriched in 543 GO terms and 44 KEGG pathways,of which Gn RH signaling pathway was the most significant pathway,while the 9221 trans-targeting genes were significantly enriched in 825 GO terms and 83 KEGG pathways,of which the metabolic pathway was the most significant pathway.In summary,the histological structure of the testes of Lande geese at 700 days of age is more conducive to spermatogenesis,and the expression levels of the spermatogenesis-related genes in the testes were also higher in Lande geese,which may be related to the differential expression of lncRNAs which target the genes enriched in the Gn RH signaling pathway and metabolic pathway. |
