| At present,the screening of differentially expressed genes in sheep mostly focuses on a single trait of different breeds,and the RNA-Seq technology is used to combine growth performance,slaughter performance and meat quality,and then to screen and regulate growth,development and meat quality related genes.Research reports are less.In order to systematically study the differences in the growth performance,slaughter performance and meat quality of Small Tail Han sheep and Du Han F1 at the genetic level,pure breeding Small Tail Han sheep male lambs and Du Han F1 male lambs(50each)were selected as the research objects,and growth was measured.Performance,and then slaughtered(small-tailed Han sheep and Duhan F1 each randomly selected 6)to determine the comparison of slaughter performance and meat quality,Masson staining of the longissimus dorsi muscle to compare differences in muscle fibers,and transcriptome sequencing and bioinformatics Analyze to explore genetic differences.The main findings are as follows:1.DP×STH and small tail Han sheep have no significant differences in height and body oblique length(P>0.05),but the live weight and bust circumference of DP×STH before slaughter are significantly higher than those of small tail Han sheep.Sheep(P<0.01);2.There is no significant difference in the carcass net meat rate of DP×STH and small tail Han sheep(P>0.05).However,in terms of carcass weight,net meat weight,net meat rate,bone-to-flesh ratio,slaughter rate and eye muscle area,DP×STH was significantly higher than small tailed sheep,and the difference was extremely significant(P<0.01);3.The marble pattern score of DP×STH is extremely significantly higher than that of small-tailed Han sheep,and the shearing force of small-tailed Han sheep is extremely significantly higher than that of DP×STH(P<0.01).In addition,the water loss rate and meat redness of DP×STH were also higher than that of small-tailed Han sheep,and showed significant differences(P<0.05),but the two groups had no effect on the loss of cooked meat,the brightness of the meat and the yellowness of the meat.Significant differences.4.Masson statistics show that the average muscle fiber area of DP×STH is larger than that of small-tailed Han sheep(DP × STH =129.243μm2,small-tailed Han sheep=108.333μm2),but the difference is not significant(P>0.05),but Duhan hybrid sheep The diameter of single bundle of muscle fibers was significantly smaller than that of small-tailed Han sheep(P<0.05)(DP×STH = 39.986 μm,small-tailed Han sheep =43.961 μm).5.DP×STH population has a total of 21,176 annotated genes,including 20,022 known genes and 1154 new genes;small tail Han sheep population has a total of 20,884 annotated genes,19,740 known genes,and 1,141 new genes.6.A total of 874 differentially expressed genes(DEG2)were identified with Small Tail Han sheep as a control,of which 457 DEGs were down-regulated and 417 DEGs were up-regulated.These genes are significantly enriched in 653 GO items,including14 cell components,10 molecular functions and 21 biological processes.There are 14 significantly different pathways.It was found that 15 unique GO items are closely related to growth and development,and 11 are closely related to meat quality or muscle structure.It was found that 30 DEGs are closely related to growth,116 DEGs are related to development,and 32 DEGs are related to meat quality.Seven genes including FGFRL1,ITGB3,BMP2,PPARD,TNF,CAV1 and IFRD1 are co-expressed in growth,development and meat quality.7.qPCR verified the expression levels of 7 genes closely related to growth,development and meat quality,and verified the three significantly different proteins(PPARD,FGFRL1 and ITGB3)by Western blotting,q PCR and western blot verification results and transcription The group analysis results have similar trends.The study believes that the growth performance,slaughter performance and meat quality of Duhan F1 are better than small-tailed Han sheep,indicating that Duhan F1 has more economic value and should be promoted.Through GO annotation and KEGG enrichment,the differentially expressed genes were analyzed,and the key genes closely related to growth,development and meat quality were further screened out,providing a scientific basis for improving sheep breeding. |
