Jiashi melon(Cucumis melo)is a Cucurbitaceous crop,mainly planted in Jiashi County,Kashi City,southern Xinjiang.It has a history of more than 1,500 years of cultivation and is a treasure in Xinjiang.As a characteristic melon crop in southern Xinjiang,Jiashi melon is of great significance to the economic and social development of southern Xinjiang and the continuous increase of agricultural income.In the production of Jiashi melon,fertilizer and virus disease are two important factors that restrict the yield and quality of Jiashi melon.According to the field production requirements of Jiashi melon,this study conducted preliminary investigation and sampling of virus disease of Jiashi melon in Jiashi County,Kashi City,southern Xinjiang,and conducted molecular mutation analysis of the Watermelon mosaic virus(WMV)that infects Jiashi melon.A preliminary analysis of the biological characteristics of Aphis gossypii,an important vector of cucurbit virus diseases,was performed.In addition,the allelopathy of various legumes such as bitter beans and other legumes on Jiashi melon was studied,in order to provide a basis for the prevention and control of virus disease of Jiashi melon,the development and utilization of bitter beans resources,and the development of green manure of Jiashi melon.The main results obtained in this study are as follows:1.This study was conducted in 4 townships in Jiashi County,Kashi City,southern Xinjiang in 2019,which have a large area of cultivation,namely,Wolituogelake Township,Guleluke Township,Kezilesu Township,and Kezileboyi Township.Investigation and sampling of virus disease of Jiashi melon were carried out,and 57 samples were collected.The results showed that the virus disease is widely distributed in the main Jiashi melon planting areas in Jiashi County and the symptoms are more diverse.The virus disease has become one of the main diseases that endanger Jiashi melon in production.In the field,virus disease mainly manifests symptoms such as mosaic,blister,deformity and leaf yellowing.Viral diseases seriously affect the normal growth and development of Jiashi melon,weaken the flavor and reduce sugar content of Jiashi melon,and seriously affect the yield and quality of Jiashi melon.2.Two-step RT-PCR was used to amplify the coat protein(CP)gene of WMV from the collected samples of virus disease of Jiashi melon,and the full-length cp gene sequence of WMV was obtained by gene cloning and sequencing.Molecular mutation analysis was performed.The results showed that the WMV isolate obtained in this study is located on clade A.The similarity comparison results showed that the sequence of the cp gene has the highest sequence similarity with the two WMV isolates WCJ-2 and WCJ-8 from Xinjiang,reaching 98.3% and 98.0%,respectively.3.To investigate the biological characteristics of Aphis gossypii in Kashi City,this study is the first analysis of the effects on the growth of Aphis gossypii under illumination and the effects on the growth of pumpkin.The results showed that when the illumination time was 5 days,the illumination had a certain effect of stimulating the reproduction of Aphis gossypii.After 5 days of illumination,the illumination had a certain effect of inhibiting the reproduction of Aphis gossypii.It had a certain influence on the growth of pumpkin plants.Total height increment of pumpkin plants in the control group reached 11.00 cm,while the total height increment of plants in the treatment groups(pots 1 to 5)was smaller than that of the control group,The increments were 10.0 cm for No.1,10.50 cm for No.2,and 6.50 cm for No.3.The number 4 pot was 8.30 cm,and the number 5 pot was 9.90 cm.Therefore,light can affect the reproduction of Aphis gossypii in Kashi City in southern Xinjiang.In addition,Aphis gossypii can affect the growth of pumpkin plants.4.In order to explore the effects of Sophora alopecuroides on the seed germination and plant growth of Jiashi melon,this study adopted the culture dish filter paper method and potted method,and analyzed the effects of different organs of Sophora alopecuroides on the seed germination and seedlings of Jiashi melon through the determination of morphological and physiological and biochemical indicators.The influence of plant growth is expected to provide a basis for the cultivation of Jiashi melon and the protection and utilization of bitter bean resources.The results showed that the leaves,stems,pods and seed extracts of Sophora alopecuroides all had inhibitory effects on the germination of Jiashi melon seeds.Among them,the pod extract had the strongest inhibitory effect and the leaf extract had the weakest inhibitory effect.The extracts of leaves,stems,pods and seeds of Sophora alopecuroides can promote the growth of Jiashi melon seedlings,and the extracts of stems can promote the growth of seedlings.In addition,the leaves,stems,pods,and dry seed powder of Sophora alopecuroides can promote the plant height,thickness,total leaf area,dry matter accumulation(dry weight)and chlorophyll content of Jiashi melon.Among them,the dry seed powder promotes the strongest effect.5.By comparing the effects of Leguminosae seed dry powder leguminous plants on seed germination and plant growth in outdoor pot,the leguminous plants with less inhibitory effect on seed germination and better promoting effect on plant growth were screened out,so as to provide basis for the development of green fertilizer of bitter bean.With Jiashi melon as the test crop,six legumes,including Sophora alopecuroides,Trifolium repens,Medicago sativa,Pisum sativum,Vigna unguiculata and Vigna radiata,were selected as test materials to study the effects of different Leguminosae seed dry powder on the germination of Jiashi melon seeds and Jiashi melon plants.The influence of morphology and related physiological and biochemical indicators.Dry seed powders of 6 legumes have inhibitory effects on the germination of Jiashi melon seeds,among which clover seeds have the strongest inhibitory effects and Vigna radiata seeds have the weakest inhibitory effects.Sophora alopecuroides seed powder has a higher promoting effect on the plant height,leaf area,dry weight and chlorophyll content of Jiashi melon than other legume seeds.Compared with other leguminous plants,the dry seed powder of Sophora alopecuroides has a better promotion effect on the plant morphology and related physiological and biochemical indexes of Jiashi melon.Therefore,in the development of Jiashi melon green manure,Sophora alopecuroides seeds can be selected as a priority.
