| As the use of growth-promoting antibiotics is further restricted,it is particularly important to develop new antibiotic alternatives to promote growth of livestock and poultry.Traditional Chinese medicine has the advantages of low cost,environmental friendliness,and resistance to drug resistance.It is one of the directions worth exploring in the field of growth promoters for livestock and poultry.The compound Chinese veterinary medicine "Zhenqi Granules"(ZQG)developed by our laboratory in cooperation with the company in the early stage uses Ligustrum lucidum and Astragalus as raw materials.Preliminary experimental results show that the compound Chinese veterinary medicine has a significant effect on promoting pig growth.In order to evaluate the safety of the Chinese veterinary drug preparation and clarify its possible adverse reactions,this study carried out acute toxicity and subacute toxicity tests.At the same time,the effect of the compound Chinese veterinary medicine on the levels of growth-related hormones in mice was preliminarily explored.The main results obtained are as follows:In the acute toxicity test,mice were given ZQG solution with different dosages(0~50 mg/10g·bw)by intragastric administration,all the mice were still alive.So,the maximum tolerance test was conducted at 50mg/10g·bw dose,and the results showed that all the mice had no death and abnormal performance.The maximum tolerance of ZQG on mice was greater than 50mg/10 g.bw.According to the "Technical Guidelines for Acute Toxicity Studies of Chemical Drugs",the entire acute toxicity test can be ended if the mice have not died at a dose of 50 mg/10 g·bw.In the subacute toxicity study,according to the Chinese Veterinary Pharmacopoeia and the "Methodology of Traditional Chinese Medicine Pharmacology" compiled by Chen Qi and others,for new veterinary drugs with a clinical course of less than two weeks,choosing a subacute toxicity cycle of one month can be well supported.One-step clinical trial.Because the clinical course of ZQG is within two weeks,we choose a 30-day administration cycler.Rats were given the drug by gavage at high,medium and low doses(50.0,25.0 and 12.5 mg/10g·bw)once a day for 30 days.The results showed that there were no abnormalities in behavior,spirit,feeding,drinking water,feces and body weight in the experimental group of rats treated with high,medium and low dose compound ZQG compared with the control group.At the end of the experiment,the blood routine indexes and blood biochemical indexes of the rats were tested,and the results showed that there were a few statistical differences between the ZQG treated rats and the control rats.However,except for the high dose group,the indexes of glutamate transaminase were higher than the normal reference range.After continuous administration for 15 and 30 days,the viscera coefficients of each group were determined,and the results showed that there was no significant difference between the rats in the administration group and the blank control group,and no pathological changes related to the effects of the drug were found in the histopathological examination.In the experiment to evaluate the effect of ZQG on the growth hormone level of mice,three dose groups(3.40,1.70,0.85mg/10g·bw)of ZQG(3.40,1.70,0.85mg/10g·bw)and Wuweijianpi granule(WJP)group(5.10)were set.mg/10g·bw),and five test groups as a blank control group(equal volume of saline).Gavage twice a day for 7 days.On the 3rd and 7th day of administration and on the 3rd and 7th day after drug withdrawal,the weight and food intake of mice in each dose group were weighed and recorded.On the 7th day of administration and the 7th day of drug withdrawal,half of the mice in each group were killed respectively,and the organs were separated and the organ coefficients were measured.The serum was taken to determine the contents of the four hormones of GH,T3,T4 and IGF-1.Analysis of the significance of the difference.The results showed that the average body weight,daily gain,and daily feed intake of the mice in each dose group of ZQG were greater than those of the blank control group in the 7 days of administration and 7 days of drug withdrawal,but there was no significant difference between the two groups.Sexual difference(p>0.05).There were no significant differences in the organ coefficients of the heart,liver,spleen,lung and kidney of the mice in each dose group of ZQG and the WJP group and the blank group(p>0.05).The results of hormone determination in mice showed that on the 7th day of administration,the contents of the four hormones in the serum of the mice in each dose group of ZQG were significantly greater than that of the blank control group(p<0.05);The levels of the four hormones in the serum of mice in each dose group of ZQG were also significantly higher than those of the blank control group(p <0.05).Based on the above results,ZQG has no obvious acute and subacute toxic effects,and can increase the hormone levels of GH,T3,T4,and IGF-1 in the serum of mice. |
