| Lentinula edodes is a kind of famous medicinal fungus,has an important status in the edible fungus industry in China.With mushroom industry development and factory production rise gradually,the traditional solid strain production pattern due to the long production period difficult to adapt to the requirements of the large scale and the anniversary of the production of edible fungi.The liquid spawn has some characteristics with a short production cycle,high purity,low rate of pollution and the high degree of standardization.This study choice the cultivated varieties L808 which cultivation area widely were used as a tested strain.The L.edodes liquid fermentation medium is optimized,was determinated the mycelium growth curve,and then use the optimized culture medium for L.edodes liquid spawn preparation and cultivation test.This study adopts the single factor experiment to determine the medium carbon source is glucose,corn flour,fine wood chips,nitrogen source is soybean meal.Single factor experiment found the wood leaching solution was promoted the Lentinula edodes spawn growth.Based on orthogonal experiment,the optimal culture medium were obtained as glucose 2%,corn flour 2%,fine sawdust 1.5%,1.2% soybean meal,medium C/N ratio of23.6:1,before the mycelial dry weight is increased by 307%.To determination of medium growth curve,the end point was fermented for 11 days;The study found that fine wood chips on the dry weight of mycelium,mycelial growth rate,the density of fungus ball has significant influence.The various characters during L.edodes liquid spawn fermentation process of correlation analysis showed that the mycelial dry weight was significantly correlated with mycelial pellets diameter,density,mycelial pellets density and diameter were significantly related.The evaluation content,evaluation method and evaluation parameters research of Lentinula edodes liquid spawn shows that the quality qualification standards for mycelial pellets diameter < 1200μm,the mycelial pellets density of 50±5 a/m L,fermented liquid p H < 3.2,the strains on the cultivation of material 24 h can germinate and lock joint was observed under a microscope.This study shows that the liquid strains of inoculation and cultivation of best solidliquid ratio(W/V)of 37.5:1-40:1.In autumn trial,vaccination 7 ml liquid bacteria pollution rate up to 18%,inoculation amount for 12 ml and 17 ml pollution rate is only 2%,lower than solid strains pollution rate(10%).The 7 ml,12 ml inoculated quantity and 17 ml,hyphae bags full time earlier than solid strains respectively 6.9 d,5.1 d and 4.6 d,reached significant difference levels.In the liquid spawn the spring and autumn cultivation experiment,inoculated liquid spawn and solid strains,the fruiting body yield was no significant difference.Add a certain amount of lime and gypsum in cultivation matrix,observe the add quantity influence for liquid strains germination and growth,the influence of the results found that the higher the concentration of lime,can inhibit the growth of the sprout hypha,gypsum concentration from 0.5% to 2% and there was no significant effect on mycelial growth. |
