| As an important platform for trade and trade exchanges between China and ASEAN countries,CAEXPO played an important role in promoting the economic development of Guangxi.With the increasing of China’s foreign trade year by year,higher requirement have been put forward for the facilitation of customs clearance in international exhibitions.The current traditional mode of government supervision of CAEXPO was not in line with its development requirements.At present,the identification of plant quarantine insects was mainly morphological method during international exhibition,but the early morphological and incomplete insects of the insects were difficult to be identified,the results were subjective,which could affected the accuracy of quarantine work and the detection rate of the epidemic.DNA Barcoding was a new technique of biological classification and identification in recent years.It has been widely used in the research of species identification,genetic evolution and so on.In this study,the technique was applied to the identification of quarantine pests carried by exhibits in CAEXPO,the main research results were as follows:1.From 2004 to 2019,total intercepted 119 species of pests in CAEXPO,more than 2,270 species,of which insects and species were the most numerous,with more than 2,225 species were intercepted,the proportion was as high as97.84%,followed by weeds,with more than 41 species were intercepted,with 8species has been identified,the proportion was 1.81%.The highest number of harmful species intercepted was 599 in 2012,and the highest number of harmful species intercepted was 35 in 2015.The highest number of pest species intercepted was 68.44% in Indonesia,and the highest number of exotic species intercepted were 54 species in Indonesia and 55 species in Cambodia.In the past15 years of the CAEXPO,10 species of quarantine pests were intercepted,including 1 species of weeds,7 species of insects,and the most frequently intercepted exotic organisms.Coleoptera(23 families,2022 species)was the largest number of exotic insects intercepted in all previous CAEXPO.2.The digital specimen library of pests has been preliminarily constructed,which included more than 140 kinds of cross-border exhibits and packages of harmful insects,more than 80 kinds of harmful insects,among which 35 kinds have been newly collected and identified,and more than 500 typical symptoms of insect hazards and more than 360 images of insect morphology have been taken.In addition,the date base of the digital platform of pest harm symptom and insect specimen library has been preliminarily built.3.Based on the constructed databaseof pests intercepted in cross-border exhibits in CAEXPO,and combined with Nanjing customs and Nanjing Herring technology Ltd.,we updated and improved the image identification system of harmful organisms/APP.With this APP,we identified the insects on rice and corn intercepted from CAEXPO.Four kinds of storage pests were identified,such as Oryzaephilus surinamensis(Linnaeus),Tribolium castaneum(Herbst),Sinoxylon anale(Lesne),Callosobruchus maculatus(Fabricius).Meanwhile,in order to realize the rapid detection,we designed the specific primers and constructed dubplex PCR system to quickly detect and distinguish S.analeand S.conigerum.The sensitivity of the system was tested,and the results showed that the template concentration of 0.05ng/μL could be detected by the dubplex PCR system and to realized the rapid detection of pests. |
