Moso bamboo(Phyllostachys edulis),with the largest planting area and the widest distribution in China,is the most important bamboo species used for eating shoots.The content of cyanogenic glucoside increased and the bitter taste increased,which restricted the development of fresh bamboo shoots industry.In the production practice,the bitter taste can be reduced by covering soil and avoiding light,but this operation takes time and effort.With the completion of genome sequencing of moso bamboo,more and more molecular studies on bamboos have been conducted,and more key genes of metabolic pathways have been screened out.Transcriptome sequencing was used to analyze the effects of light-avoidance treatment on cyanogenic glucoside and related metabolic pathways of moso bamboo,which will provide theoretical guidance for the development of moso bamboo and other bamboo species.In this paper,the effects of soil covering and shoot emergence on the content of cyanide and cyanogenic glucoside in Bamboo shoots and transcriptome sequencing were carried out.The results showed that:The cyanogenic glucoside in the bamboo shoots was in the form of linamarin and lotaustralin,the content of lotaustralin was higher than linamarin,and the content of linamarin and lotaustralin was about the same as that of cyanide.With the growth direction of moso bamboo shoots,the content of cyanide,linamarin and lotaustralin increased gradually(basal <middle < apical),and the content of the three substances was the most significant to reduce the apical of bamboo shoots by avoiding light.With p-adjust < 0.05 and log2│FC(EP/CK or US/AS)│≥1,1074 differentially expressed genes were screened out of the transcriptome sequencing data of CK and EP.Compared with CK in the control group,827 differentially expressed genes were up-regulated and 247 were down-regulated.The differentially expressed genes of formamidase[EC:3.5.1.49] related to the synthesis and degradation of cyanide,linamarin and lotaustralin were screened in the Map00460 Cyanoamino acid metabolism pathway:PH01000099G0750 and PH01000625G0150,in which PH01000099G0750 was up-regulated in EP and PH01000625G0150 was down-regulated in EP.767 differentially expressed genes were screened out of the transcriptome sequencing data of AS and US.Compared with the AS control group,365 differentially expressed genes were up-regulated and 402 were down-regulated.The differentially expressed β-glucoside[EC3.2.1.21] genes related to the synthesis and degradation of cyanide,linamarin and lotaustralin were screened in the Map00460 Cyanoamino acid metabolism pathway:PH01000027G1630 and PH01000625G0150,in which PH01000027G1630 was down-regulated in US and PH01000625G0150 was up-regulated in US.
