| Ostrya rehderiana Chun,a perennial woody plant belonging to the Betulaceae family and the genus Ostrya,is a rare and endangered plant unique to China.Only 5 native plants are distributed in the West Tianmu Mountain of Zhejiang Province and have been listed by the country Wild plants are protected as first-level key protection.Ostrya rehderiana is of great value in economics and scientific research.In this study,the transcriptome of the leaves of Ostrya rehderiana was sequenced,EST-SSR molecular markers were developed,genetic analysis and parental analysis of Ostrya rehderiana introduced populations in Tianmu Mountain,Qingyuan,Wuchao Mountain and other places were carried out,In order to provide a theoretical reference for the protection and propagation of Ostrya rehderiana.The conclusions are as follows:(1)The high-throughput sequencing technology is used to sequence the Ostrya rehderiana transcriptome and analyze the sequencing data,which enriches the data of Ostrya rehderiana gene.At the same time,EST-SSR site discovery and the development of corresponding primers were carried out.Finally 12 pairs of primers with high polymorphism were obtained,which filled the vacancy of the SSR molecular marker of Ostrya rehderiana.(2)The geographical location and growth environment of each introduction area are different,but the Ostrya rehderiana grows well in each introduced area,indicating that Ostrya rehderiana still has strong adaptability,can survive and grow well in different environments.(3)From the results of genetic diversity testing,the degree of genetic diversity is not high but it does not show very low genetic diversity.There used to be more ancestral populations around the original plants of Ostrya rehderiana.May be the reason why only 5 original plants and their progeny still retain relatively high genetic diversity.(4)In the comparison of the genetic diversity of different introduced populations,it is found that from the population perspective,the genetic diversity of the introduced population is higher than that of the original plant population,but from the individual perspective,the genetic diversity of the introduced individuals is The original plant is lower.(5)According to the results of parental analysis,the genetic differences caused by the parents for each introduction site are not obvious.Based on the above conclusions,the following protection strategies are proposed:(1)Strengthen the conservation of original plants and the preservation of genotypes;(2)Collect seeds from various introduced populations for propagation and resource conservation;(3)Introduce species in different habitats,And analyze the environmental factors that restrict its growth;(4)Deepen the research on the genetic diversity and endangerment system of Ostrya rehderiana.(5)Increase publicity and strengthen people’s awareness of the protection of endangered plants. |
