Konjac belongs to Amorphophallus Blume of Araceae,with tropical rainforest climate as the main origin and widely distributed in Southeast Asia,Africa and other countries.China is the mainly distribution of konjac,and the konjac species discovered account for about 1 / 10 of the total number of konjac in the world,among which more than 10 species such as A.albus and A.sinensis are unique to China.In this paper,the genetic diversity of konjac germplasm accessions was conducted to better understand their relationships and identify quality konjac germplasm resources from the aspects of phenotypic diversity,DNA barcoding and chromosome karyotype diversity.The results will promote the sustainable development of konjac planting industry.The main results are as follows:1.Based on 17 morphological characteristics,the leaf types of 60 konjac plants were studied.The samples were divided into 4 groups by cluster analysis.The results showed that the genetic variation of each konjac sample was obvious,the average coefficient of variation of leaf type was 93 %,among the coefficient of variation of each phenotypic character,whether there were bulbils at the fork of petiole > t he smoothness of petiole > the shape of leaf tip.2.Based on the matK sequence,DNA barcode technology was used to amplify the konjac materials.The NJ phylogenetic tree was constructed for the 41 sequences successfully amplified,which were divided into 7 categories.Compared with the traditional morphology,the clustering results of most materials are basically consistent,and there are some differences in the classification results of a few materials,such as A-19-10,A-19-26,A-19-28,A-19-53 and A-19-47 in the phylogenetic tree.3.In this study,the number of three types of chromosome was analyzed,among which26,28 and 39 samples accounted for 56.7 %,5 % and 38.3 % of the total samples respectively.Among them,A-19-18,A-19-25 and A-19-31 are consistent with the results of morphological classification.The chromosome types of this group of materials arerich,and the biggest characteristic of morphological characteristics is that the surface of petiole is rough with verrucous grain,belonging to the A.virosus;meanwhile,the triploid plants with chromosome number of 39 have strong and strong resistance.According to the characters such as the color of petioles,the color of lobes,and the glossiness of leaves,A-19-3 was identified as A.bulbifer;A-19-14 as Gengyu 2;A-19-15 as A.muelleri,consistent with the results of DNA barcode analysis;A-19-16 as A.krausei;A-19-21 as A.konjac,and A-19-29 as A.yunnanensis.
