| Anthurium andraeanum Linden is a rare tropical flower belonging to the genus Anthurium Schott of Araceae.It can be used as potted ornamental plant or cut flower plant.Cross breeding is the main breeding method of anthurium anthurium,but the breeding of anthurium andraeanum new varieties and the development of anthurium industry have been seriously affected by the factors such as the long generation cycle of anthurium andraeanum,the great difference of fertility between different varieties,and the unclear genetic mechanism of fertility related traits.It is of great significance to study the genetic diversity of anthurium traits and construct the high-density genetic map to understand the genetic mechanism of anthurium and promote the development of new anthurium varieties.In this study,a genetic separation population was constructed by hybridization of "Purple Princess" of "Tulip" type and "Semper" of "Mickey" type,to draw high-density genetic map of anoparpus,and to analyze the genetic diversity of main morphological traits of the mapping population.The results show that:(1)according to the breeding objectives,anthurium varieties with different Buddha flame bract shapes are selected directionally as parents,and hybrid populations are arranged in a planned way.Finally,"Tulip"(Purple Princess)and "Mickey"(Semper)with distinct phenotypic traits were determined as the parents of F1 generation population that constructed the high-density genetic linkage map.40 plants of each variety were selected for mass hybridization to obtain F1 generation,and 150 individuals were randomly selected to form the mapping population.(2)16 main morphological traits of the mapping population were investigated and counted.The results showed that all the morphological traits in F1 generation were widely separated,showing continuous variation and obvious heterosis,with the coefficient of variation ranging from 15.43% to 115.08%.The results of correlation analysis showed that the majority of the main morphological characters of anthurium were positively correlated,and the morphological characters of foyanbract were significantly positively correlated.Three principal components were selected by principal component analysis,which were determined by crown width,bud shape and inflorescence thickness.Cluster analysis groups can be divided into three groups,including the first Ⅱ groups better,has the potential application in the subsequent individual fine screening.(3)simplified genome sequencing was performed on the two parents and 150 progeny of the mapping population,and a total of 288,404 SLAF tags were developed,of which115,973 were SLAF tags with polymorphism and 46,353 were SLAF tags with polymorphism.The average parental detection depth in the mark above is 86.86 x and21.37 x.Then,the developed markers were genotyped to obtain 9,295 markers in the figure above.The total distance of the constructed high-density genetic map was 2,742.49 cm,the integrity of the markers in the figure above was 99.95%,and the average distance of the map was 0.40 cm,containing a total of 15 linkage groups.In this study,anthurium hybrid F1 generation population was used as the mapping population to analyze the genetic diversity of the main morphological traits of the mapping group,find the correlation between different traits,dig out the three principal components that affect the diversity,and cluster them successfully.It provides references for the selection of good traits,the selection of high quality anthuriums parents,the research of crossbreeding and the understanding of the genetic mechanism of anthuriums.The high-density genetic map of anthurium was constructed and high-quality genetic map was obtained,which laid a foundation for the subsequent QTL mapping and the acquisition of a large number of QTL loci and primary QTLS controlling major traits,and the understanding of the internal genetic mechanism of anthurium from the gene level. |
