| Soft corals are symbiotic with zooxanthellae,bacteria,and fungi,and have strong wound self-healing capabilities.Related research shows that they have potential biomedical applications.In this study,Sarcophyton trocheliophorum,the dominant species around Hainan Island,was used as the material.High-throughput sequencing technology was used to compare and analyze the transcriptomes of the samples of pre-traumatic and24-hour post-traumatic soft corals.The oxidoreductase activity on the 1st,3rd,7th,15 th,and 30 th days after trauma,thus preliminary revealing the molecular mechanism of soft coral wound repair.The main findings are as follows:1 Through transcriptome sequencing,a total of 258,677,462 reads were obtained for the six samples,145,175 genes were obtained after software assembly,and 47,614 annotated genes were obtained through nine database comparisons.Among them,37,477 genes were annotated into the Nr database,35,369 genes were annotated into the GO database,and 31,786 genes were annotated into the Swissprot database.These gene sequences enriched the Hainan soft coral transcript database.2 Analysis of differentially expressed genes in the trauma group and the control group showed a significant difference in 151 genes,of which 134 genes were up-regulated and 17 genes were down-regulated.Functional enrichment analysis of differentially expressed genes,oxidation-reduction process(GO0055114),oxidoreductase activity(GO0016491),photosynthesis(GO0015979),photosynthetic system(GO0009521),photosynthetic system II(GO0009523),and photosynthetic electron transfer(GO0009767)Functions get enriched.3 Randomly select 5 differentially expressed genes and verify the coral transcriptome data by fluorescence quantitative PCR.The results showed that the expression level of 3genes was significantly up-regulated and the expression level of 2 genes was significantly down-regulated,which was consistent with the transcriptome data,indicating that the analysis of transcriptome gene sequence information and expression was accurate.4 Enzyme activity indicators showed that oxidative stress occurred in Sarcophyton trocheliophorum after human trauma,and SOD activity increased about 6 times after 24 hours of trauma,reaching a peak(122.56 ± 29.36)U / mg prot,and gradually decreased until the seventh CAT activity recovered to the same level before trauma;CAT activity increased about 4 times after 24 hours of trauma,reached a peak(13.40 ± 3.89)U / mg prot,and then gradually decreased until the same level before trauma on the 7th day,while rosette meat The GST activity and LPO content of Sarcophyton trocheliophorum increased significantly in a short time after trauma.In addition,the determination of coral symbiotic algae density and chlorophyll a content found that within a short period of time after the coral was injured,corals would emit part of the zooxanthellae stress response,and the change in chlorophyll a content would lag behind the change in symbiotic algae content. |
