| Limited by growing environment,plants suffer from abiotic stress in the process of evolution and growth.As a kind of economic plants,growth and development of horticultural plants are influenced by abiotic stress,such as drought,cold,salt etc,which affect economic benefits a lot in turn.Pyrus,one of the most important economic horticultural plants,endures drought effection for many years.Hwangkumbae,belongs to Pyrus pyrifolia,loved by many people around the world,lacks drought resistance ability,wihch makes Hwangkumbae being infringed easily by drought stress.After years of research results,metabolic pathways of plant under drought stress has made significant breakthrough.Among them,the interactions of each gene in ABA metabolic pathways have been gradually clear.It has more research on various species,however less in Pyrus.By manual processing to Hwangkumbae and comparing of the transcriptome data analysis,three key genes of ABA’s metabolic signal conduction were selected.After cloning the gene,analyzing their sequence,qRT-PCR experiments,constructing agrobacterium vectors,and getting transgenic plants,some results were gotten as follows:(1)The full length of PpPP2 C is 1138 bp,387 amino acids were coded.The full length of PpSnRK2 is 1024 bp,340 amino acids were coded.The full length of PpABI5 is 1297 bp,431amino acids were coded.(2)Relative expression of PpPP2 C increases with the increase of drought days and achieves maximum value after returned to normal growth conditions.Relative expression of PpSnRK2 decreases with the increase of drought days and achieves maximum value after returned to normal growth conditions.Relative expression of PpABI5 increases with the increase of drought days and achieves maximum value after returned to normal growth conditions.(3)This three gene recombinant pCAMBIA1301 vectors were successfully built.And they have been transferred into model plant arabidopsis thaliana,Homozygous generation screening process is under way. |
