Study On The Content Of Polyphenols And Their Metabolism-related Enzyme Activities In Walnut

In this study,walnut varieties of ‘Zhonglin no.5’,‘Xiangling’,‘Gifting no.1’ and ‘Gifting no.2’ were used as test materials to study the dynamic changes of polyphenols contents and related enzyme activities in different parts of walnut leaves and fruits at different phenological stages:1.The results showed that 12 kinds,8 kinds,10 kinds and 12 kinds of monomer phenolic substances were detected in the leaves,seed kernel,seed coat and green peel of 4 walnut varieties.The monomeric phenolic substances with the highest content in different organs of the four varieties were different.For example,rutin was the monomeric phenolic substance with the highest content in leaves of "gifting no.2" and "zhonglin no.5",while catechin was the monomeric phenolic substance with the highest content in leaves of "xiangling" and "gifting no.1".The monomer phenols with the highest content in seed kernel,seed coat and green peel were caffeic acid,rutin and walnut quinone.In addition,the monomer phenolic substance also has the significant variety difference.2.In the different stages of different varieties of walnut leaves polyphenol components are analyzed,and the results show that the ’gift 1’,’sweet ling gift’ and ’2’ blade catechins content in the overall rising with the advance of developmental stage,and ’eng.5,catechins content in the leaves’ down-up-down of change process;Except for "gift no.1",the content of rutin increased first and then decreased during the development of several varieties.In addition,the dynamic accumulation of other monomer phenolic substances also has significant differences in varieties.3.In the different stages of different varieties of walnut leaves and green husk polyphenols metabolic enzyme activities were analyzed,the results showed that the four varieties of leaf in DAHPS、CHS、CS and CM activity in the late development is higher,the activity of PAL solution in the whole development stage were relatively stable;DAHPS activity in fruit green skin of four cultivars was higher than that in leaves during development,and CHS activity in green skin was higher in the early stage of development.The change pattern of CM activity in green skin of four cultivars was similar,and the pattern was "up-down-up-up".The PAL activity of the four cultivars was relatively stable in the whole development stage,which was consistent with the change rule of PAL activity in leaves.4.Components of polyphenols in different varieties and green husk and metabolic enzyme activity carried on the correlation analysis,the results showed that the four varieties of participation shikimic acid synthesis pathway in DAHPS activity,CS and CM respectively with Yang mei element,the highest correlation coefficient,chlorogenic acid and coffee acid in benzene propane way C4 H and CHS and the activity of PAL respectively with of coumaric acid,caffeic acid and coumaric acid the highest correlation coefficient,however the metabolic enzyme activity in the leaves in the correlation between polyphenol composition has not been reached significant level;DAHPS of walnut green husk,the activity of the CS and CM,respectively,and ferulic acid,chlorogenic acid and highest correlation coefficients of coumaric acid,C4 H in green husk,CHS and the activity of PAL and rutin,Yang mei,and chlorogenic acid respectively the highest correlation coefficient,the correlation between components and parts and enzyme activity reached significant level,such as green husk of ferulic acid significant correlation with DAHPS activity and activity of coumaric acid and CM significant negative correlation.5.The results showed that the activities of polyphenolic metabolic enzymes in walnut varieties were positively correlated,and most of them reached a significant level.DAHPS was positively correlated with C4 H,CHS and CM.There was a significant positive correlation between C4 H and CHS,CS and CM.CHS was significantly positively correlated with CM and CS.
