| This experiment uses Spathiphyllum,Chrysanthemum,Dendrobium chryseum and Dendrobium officinale sarcophagus as test materials,using violet light(UV-A),blue light(B),green light(G),yellow light(Y),red light(R),and far-red light(Fr)of LED light sources.Designing 12 different light qualities(UV-A,B,G,Y,R,Fr,80%R+20%B,70%R+30%B,60%R+40%B,80% R+20%B+Fr,70%R+30%B+Fr,60%R+40%B+Fr),using ordinary fluorescent light as a control(CK),exploring different light qualities for tissue culture and hydroponics The effects of growth and development were screened for LED light sources suitable for Spathiphyllum cluster shoots,Chrysanthemum organogenesis,Dendrobium chryseum and Dendrobium officinale plant tissue culture,and water culture of Spathiphyllum,Chrysanthemum,and Dendrobium officinale,obtained from the tissue culture stage to the water culture.The precise LED required for the whole process of the stage provides a theoretical basis for the research and development of plant-cultured and hydroponic-specific LED light sources.The main results are as follows:1.The effect of different light qualities on the growth and development of Spathiphylluma.Experiments on the effect of different light qualities on the proliferation of Spathiphyllum tissue culture seedlings showed that the white palm multiplication coefficient was better than that of the fluorescent control with red light treatment,and red light promoted the increase of its proliferation coefficient;fresh mass,soluble protein,chlorophyll b and the like.The content of carotene was the highest under red-blue mixed light,red-blue mixed light promoted pigment synthesis,photosynthetic product and biomass accumulation,and was best treated with 80%R+20% B+Fr.Therefore,80%R+20%B+Fr is the best light source suitable for proliferation of Spathiphyllum.b.Experiments on the effect of different light qualities on rooting of Spathiphyllum showed that the plant height and SOD activity of Spathiphyllum were the highest in UV-A treatment,and purple light was beneficial to the elongation of stems and the increase of SOD activity;MDA content in the CK treatment,the fluorescent lamp increased membrane lipid peroxidation injury;leaf number and root activity in the 60%R+40%B treatment maximum;root length and fresh weight in the70%R+30%B+Fr treatment maximum,chlorophyll a,The activities of chlorophyll a+b,carotenoids,soluble sugar and CAT were the highest at 60%R+40%B+Fr;red-blue mixed light promoted leaf and root development,pigment synthesis,organic accumulation and biomass increase,and membrane system.Stable and best handled with 60% R+40% B+Fr.Therefore,60%R+40%B+Fr is the best light source for the rooting of Spathiphyllum tissue culture seedlings.c.Experiments on the effect of different light qualities on the growth and development of Spathiphyllum in water culture showed that the plant height,leaf length and fresh quality of Spathiphyllum were the highest in B treatments,and blue light promoted its stem elongation and biomass accumulation;chlorophyll a,chlorophyll b,The content of chlorophyll a+b and soluble protein in stem were the highest in R treatment.The red light promoted the synthesis of chlorophyll and protein in stem.The soluble sugar in stem and root was the highest in Y treatment,and the yellow light promoted the accumulation of photosynthetic product in stem and root.The contents of soluble protein,carotenoids and leaves were highest at70%R+30%B.Root length,root activity,CAT activity,net photosynthetic rate and stomatal conductance were the highest under 60%R+40%B treatment.The mixed light promoted the enhancement of photomorphogenesis,photosynthesis and antioxidant capacity,and was best treated with 70%R+30%B and 60%R+40%B.Therefore,60%R+40%B and 70%R+30%B are more suitable for the light requirements of the growth process and are the best light quality for Spathiphyllum water culture.2.The effect of different light qualities on the growth and development of Chrysanthemum.a.Effects of different light qualities on the induction of adventitious buds in Chrysanthemum leaves showed that the number of adventitious buds induced by Chrysanthemum leaves was the highest in UV-A treatment,and the number of adventitious buds increased in purple light;the activity of carotenoids,soluble protein and POD was 70%R.+30%B treatment was the largest,chlorophyll a,chlorophyll b,chlorophyll a+b,and CAT activity were the highest at 70%R+30%B+Fr treatment;soluble sugar and SOD activity was highest at 60%R+40%B+Fr treatment.Red-blue hybrid light promotes pigment synthesis,photosynthetic product accumulation,and maintains a stable antioxidant enzyme system,providing a suitable environment for organogenesis,with 70%R+30%B+Fr and 60%R+40%B+Fr The best deal.Therefore,70%R+30%B+Fr and 60%R+40%B+Fr are the best light qualities for the induction of adventitious buds from Chrysanthemum leaves.b.Experiments with different light qualities on the rooting of Chrysanthemum in vitro showed that plant height,root activity and POD activity of Chrysanthemum morifolium were the highest in Fr treatment,and far-red light promoted stem growth,root activity and POD activity.The leaf number and SOD activity were the highest at70%R+30%B;the contents of chlorophyll a,chlorophyll b and chlorophyll a+b were highest at 60%R+40%B+Fr;leaf length,root length,fresh weight,dry weight,carotenoids,soluble protein,and CAT activity were the highest at 60% R+40% B;red-blue mixed light promoted leaf and root development,pigment synthesis,biomass accumulation,and gave it a more sophisticated antioxidant enzyme system.The best treatment is 60% R+40% B.Therefore,60%R+40%B is the best light quality suitable for the rooting of Chrysanthemum tissue culture seedlings.c.Experiments on the effect of different light qualities on the growth and development of Chrysanthemum hydroponics showed that the plant height,fresh weight,and soluble protein of Chrysanthemum were the largest in B treatment.Blue light promoted stem elongation,biomass accumulation,and soluble protein synthesis;leaf width root activity and carotenoids were the highest in R treatment,red light promoted the increase of root activity and carotenoid synthesis;POD activity,CAT activity and intercellular CO2 concentration were the highest in Fr;leaf length and development of Dendrobium nobile.a.Experiments on the effect of different light qualities on the growth and development of tissue culture seedlings of Dendrobium chryseum showed that the plant height of tissue culture seedlings of Dendrobium chryseum was the highest in Fr treatment,far-red light was beneficial to the elongation of stems,and the root length and fresh weight were G treatment in stems.SOD activity has a promoting effect;soluble protein and soluble sugar in roots are the highest in R treatment,red light is favorable to the accumulation of carbon and nitrogen compounds in leaves,60%R+40%B and 70%R+30%B are beneficial to the water culture of Spathiphyllum,and the mixed red-blue light is conducive to the growth of tissue culture seedlings of Dendrobium officinale. |
