| The study is aimed to measure the production performance and egg quality used to clone the full length cDNA of transforming growth factor-βgene.The sequence of gene and protein encoded were analysed by using a variety of bioinformatics softwares.By PCR technology method to detect the single nucleotide polymorphism of transforming grality of the captivity Jin Ding duck.Mathematical curve models were used to fit the egg production curve of the duck.And respectively analyze the correlation on the behavior of each egg quality.Reverse transcription polymerase chain reaction(RT-PCR)and rapid amplification of cDNA ends(RACE)with transforming growth factor-β gene,and analysis of the correlation between the polymorphisms and egg quality.The results are displayed as follows:Age at found egg and age at first egg of Jinding duck was 104 days and 130 days,respectively.Egg number at 300 days was 134.65±8.32,and the average egg weight at 300 days was 75.65±5.80g.Green shell rate was 97.72%.Body weight at first egg was 1503.25±201.76g.Body weight at 300 days was 1790.46±211.83g.The egg quality result:Jin Ding duck average egg weight was 75.65±5.80g,eggshell strength was 3.923±0.756kg/cm2,egg-shape index was 1.341±0.043,shell thickness was 0.43±0.04mm,yolk color was 2.32±0.27,Protein height was 7.78±2.21,haugh unit was 82.58± 13.35.The Jin Ding duck egg quality traits of correlation analysis:eggshell strength and shell thickness was significantly positively related(P<0.05),the correlation index was 0.349;egg weight and Protein height were significantly positively related(P<0.05),the correlation index was 0.345,haugh unit and Protein height were extremely significant positive correlation(P<0.01),the correlation index was 0.967.The full-length cDNA sequence of TGF-β gene is 3225 bp in size,including a 371bp 5’Untranslated region,a 1913 bp 3’Untranslated region with the poly(A)tail(Gen Bank accession No:MG8563 83)and an open read-ing frame(OFR)of 1239 bp in length,and encodes apredicted protein of 412 amino acids.The result of sequence alignment shows that the nucleotide sequence shares 93%and 91%to Anas platyrhynchos and Gallus gallus.The amino acids sequence shares 99%identity to Gallus gallus.Phylogenetic analysis indicates that a transmembrane structure was speculated and a signal peptide was found in No.24 AA.The protein has helix of 18.20%,strand of 16.99%and loop of 64.81%in secondary structure;the three-dimensional structure of the protein was predicted.Using sequencing in captivity Jin Ding duck TGF-β gene detects a mutation(A→G)and detects the AA,AG and GG 3 kinds of genotypes.Jin Ding duck AA,AG and GG type of gene frequency were 0.24,0.36 and 0.40;A,Ggene frequency was 0.42 and 0.58 respectivel.After correlation analysis three kinds of genotypes and egg quality traits found that Jin Ding duck AA type eggshell strength was higher than that of AG and GG type eggshell(P<0.05). |
