| Picria Fel-terrae Lour has been widely used in the prevention and control of human diseases,and there are currently few reports of its use in animal disease prevention and control.The purpose of this experiment was to investigate the optimal technology for the extraction of Picria Fel-terrae Lour extracts,to establish the quality standard of the Picria Fel-terrae Lour extracts,and to add the Picria Fel-terrae Lour extracts to the diet to influence the performance of piglets.It provides experimental basis for the application of Picria Fel-terrae Lour as a medicinal additive for animal disease prevention and control in livestock and poultry production.Study on the extraction process of Picria Fel-terrae Lour:The extraction process of Picria Fel-terrae Lour was investigated.The extraction process was optimized by L9(34)orthogonal test with extraction times,extraction duration,and Alkali extraction of pH,as the main investigation factors Results:The optimal extraction conditions were as follows:Alkali extraction of pH was 11,the decoction duration was 1 h with 3 extraction times.The optimized process of Picria Fel-terrae Lour is convenient and feasible.Thin layer chromatography(TLC)method for qualitative analysis of Picria Fel-terrae Lour extracts;Using high performance liquid chromatography(HPLC)method to determine the content of piefeltarraenin IA in Picria Fel-terrae Lour extracts.TLC results showed that the Picria Fel-terrae Lour extracts and piefeltarraenin IAhave same spots in the same position with its corresponding standard herbs;HPLC results showed that piefeltarraenin IA has good linear relationship within 20.06~104.1μg/mL(R2=0.9992),the average sample recovery rate was 99.90%,RSD=0.53%(n=6).Qualitative and quantitative analysis method mentioned above were simple,the result was accurate and reliable,which could be used as the method of identification of Picria Fel-terrae Lour and content determination of piefeltarraenin IA.The extract of Picria Fel-terrae Lour was used in the diet of piglets,and the growth performance,blood biochemical,antioxidant indexes and immune factors of the three-way crossbred piglets were analyzed with different additive amounts of Picria Fel-terrae Lour extracts,The rational use of pigs in the production of piglets and the healthy breeding of piglets provide theoretical basis and production guidance.Research method:Two hundred and fifty which similar weight 28-35 days old Du*Long*Big three crossbred piglets with their body weight(no significant difference)were selected and randomly allocated into five Groups,including control group and 5 experimental groups(The groups A,B and C were high,middle and low doses of three treatment groups and fed diet with 3.5g,1.7g and 0.875g,Picria Fel-terrae Lour extracts respectively,group D was a medical control group fed a diet with 4.0g/kg Qibusan.The body weights were determined before the mixed-medium feeding,the mixed-medium feeding mid-term,and the mixed-medium feeding,and the corresponding average daily weight gain and feed-to-weight ratio were calculated.Serum samples were collected at different stages,and the effects of different doses of Picria Fel-terrae Lour extracts on the blood biochemical parameters and immune factors of the piglets were analyzed.The results showed as follows:(1)The body weight,ADG and ADFI of the piglets in the Picria Fel-terrae Lour extracts were the most obvious,and the feed-to-weight ratio was also significantly lower than that of the experimental group and the blank group.After treatment discontinuation,the weight of piglets,ADG,ADFI,and feed-to-weight ratio were still better than those of the blank control group.(2)According to the body weight,ADG,ADFI,feed-to-weight ratio and anti-oxidation immunity index of the test additive dosage group,the additive amount of 1.75 g/kg bitter genitalia extract has the best effect. |
