This paper studied the screening of the microorganism promoting tobacco mellowing and the application on tobacco aging,exploiting microbial technology applied in tobacco mellowing and find efficiently microorganism to reduce bitter and astringent of aftertaste,and degrading pectin、cellulose、protein and starch of tobacco leaf by using microbial preparation in tobacco mellowing at nature condition so as to reduce the adverse effect caused by the macromolecular、shorten the time of tobacco aging、cut down enterprise cost and improve the quality of tobacco leaf.High content of pectin、cellulose、protein and starch of tobacco leaf influence the quality of tobacco leaf and aging speed seriously.In order to degrade pectin、cellulose、protein and starch of tobacco leaf,accelerate aging,and improve quality and safety of tobacco leaf,this study isolated 123 bacterias and 9 fungi from 18 flue-cured tobacco collected from 3 age(2011、2012、2013)and 6 places of origin(Sanmenxia and Xuchang in Henan province,Nanping and Sanming in Fujian province and Chuxiong and Lincang in Yunnan province),including 38 pectin degrading bacterias、51 cellulose degrading bacterias、22 starch degrading bacterias and 18 protein degrading bacterias.Among the 38 pectin degrading bacterias,strains SMXP-58、XC-8、XC-30、SMXP-57、SMXP-62 possessed high ability of degrading pectin.Pectinase activity of them were 192、179、176、143、151 U/m L,respectively.Strain SMXP-58 possessed highest enzymatic activity(192 U/m L).Based on its morphological,physiological biochemical characters and 16 S r RNA sequence analysis,this strain was identified as Bacillus subtilis,named as Bacillus subtilis SMXP-58.Total score of smoking increased 1.02 spraying with bacteriastatic agent made from SMXP-58.Pectin of the tobacco reduced to the least 7.45% with fermenting 60 h,spraying 1m L agent per 100 g tobacco leaf,spraying 1m L water per 100 g tobacco leaf.Among the 51 cellulose degrading bacterias,strains XC-19-1、XC-19-2、XC-20、SMX-42-1、SMX-42-3、SMXP-58 possessed high ability of degrading cellulose.Celluase activity of them were 22、16、17、19、16、18 U/m L.Strain XC-19-1 possessed highest enzymatic activity(22 U/m L).Based on its morphological,physiological biochemical characters and 16 S r RNA sequence analysis,this strain was identified as Bacillus subtilis,named as Bacillus subtilis XC-19-1.Among the 22 starch degrading bacterias,strain XC-3 possessed highest enzymatic activity.Based on its morphological,physiological biochemical characters and 16 S r RNA sequence analysis,this strain was identified as Bacillus thuringiensis,named as Bacillus thuringiensis XC-3.Among the 18 protein degrading bacterias,strains SMXP-3、SMXP-58、SMP-6、NPP-10、CXP-13 、 LCP-18 possessed high ability of degrading protein.Protease activity of them were3417、2217、3246、2789、2217、3017 U/m L,respectively.Strain SMXP-3 possessed highest enzymatic activity(3417 U/m L).Based on its morphological,physiological biochemical characters and 16 S r RNA sequence analysis,this strain was identified as Bacillus punmilus,named as Bacillus punmilus SMXP-3.
