| At present,the research on artificial culture of cicada is still in the exploratory stage.Cucumber flower toxicology experiment still stays in the stage of animal experiments,the depth,whether the artificial products can replace wild products also need more experiments to demonstrate,clinical tirals involved still shallow,there is no artificial cμLtivation of high-tech products available.In this paper,the artificial cultivation methods of cordyceps cicadae were studied,and the basic analysis was carried out on the separation and detection of the relevant active ingredients,and the safety was analyzed.The main contents were as follows:1、Study on artificial cultivation and safety of the cordyceps cicadaeThe optimum growth conditions of the artificial cultivation of the cordyceps cicadae were studied.and the acute toxicity experiment was carried out to ensure the safety of the best cμLture.The growth condition was recorded by artificial clean cultivation experiment.The optimal growth environment was optimized by orthogonal test.The mice were injected with different concentrations of cordyceps cicadae suspension at one time.Tolerant dose to determine the safety of artificial cultivation of cicada flowers,observe the record the toxicity of reaction of the mice,calcμLate the maximum tolerated dose.The results showed that light,temperature and humidity had a great effect on the growth of cicada flowers.After one time,mice were injected with different concentrations of cicada suspension,and no mice were killed.Conclusion:The optimum growth environment of cordyceps cicadae was as follows:temperature 23±2℃,humidity 50±5%and culture time 40±5d.The acute toxicity test showed that the artificial cultivation of cordyceps cicadae was non-toxic and could be used as raw material for food and medicine.2、Quantitative study on the index components of artificial cordyceps cicadaeThe use of indicators of determination of content of ingredients to determine its quality standards to ensure that the cicada flowers can be safe and effective use.In this paper,quantitative determination of total sugar,adenosine,protein,fat and amino acids in the standards components was studied by phenol sulfuric acid,high performance liquid chromatography,Coomassie brilliant blue,fat analyzer and armino acid analyzer.The resμLts showed that the water content of the spore powder was 64.52±3.07%,the total sugar was 2.31±0.13%,the protein was 23.30±1.75%,the fat was16.60±0.03%;the coremium water content was 49.41±4.87%,the total sugar was 2.83±0.21%,the protein was 17.10±1.15%,the fat was 10.00±0.01%,the adenosine was 0.5911±0.098mg/g.18 kinds of amino acids were detected.There were 17 fatty acids in Cordyceps cicadae,16 fatty acids in Cordyceps militaris and Cordyceps sinensis.The content of unsaturated fatty acids(about78%)was higher than that of saturated fatty acids(about22%).The content of fatty acids in Cordyceps cicadae was higher than that of other Chinese caterpillar fungus.The content of the spore powder is slightly higher than that of the spores,which is the basis for the large-scale use of artificial cicada flowers.3、Cordyceps cicadae ethanol extract immunoassay screening testThe effects of artificial extracts of Spodoptera littoralis and its different concentrations of ethanol extract on the proliferation of splenic lymphocytes,in normal mice were observed and the effects of immune function were analyzed.Effective parts:In this study,the immunoreactive components of cicada were isolated and purified by macroporous adsorption resin column chromatography.The effects of different elution of ethanol elution on the proliferation of splenic lymphocytes induced by concanavalin were observed in different degrees.Its effect on immune function in mice.The results showed that 30%ethanol extract had the most obvious inhibitory effect on the proliferation of mouse lymphocytes stimulated by concanavalin. |
