| Ganoderma lucidum is a traditional medicinal mushroom with highly medical and nutritional values.The application in Southeast Asian countries has a long history.G.lucidum has the very good curative effects in anti-aging,improve immunity,antiviral and anticancer etc.Ganoderma triterpenes are important secondary metabolites of G.lucidum,GAs contents have become an important standard to evaluate the quality of G.lucidum.With the finish of G.lucidum genome sequencing work,the study level of G.lucidum is deepen into the level of molecular genetics from the basic biology research level,along with the establishment of a series of genetic operation platform,such as RNA interference technology and gene excessive expression system.The researches effectively promote G.lucidum become a new kind model of medicinal fungi,improving its scientific and commercial value.We use G.lucidum gene silencing system established in the laboratory to study the effect of AP1 in mycelium growth,metabolic regulation and control mechanism.At the same time,we explore the roles of the REDOX system in the GA biosynthesis in G.lucidum.The transcription factor AP1 is a regulatory factor studied earlier in fungi.AP1 is reported to regulate the corresponding genes transcription.In previous studies,the regulation of AP1 responsing to oxygen stress is the one which is fully studied.On the basis of the gene silencing system,we study the functions of AP1 on the physiological and biochemical activities including regulting the REDOX balance.At the same time,we explore H2 as a new type of exogenous signaling molecule in improving the antioxidant mechanism of G lucidum.The function analysis of AP1 transcription factor:according to G.lucidum genome sequence,we identified a homologous gene of S.cerevisiae Yap1 in G.lucidum.By comparing functional domains and evolutionary tree analysis,we confirmed it as ap1 homologous gene in G.lucidum,named apl.We used the positive promoter RNA silencing technology to study the API functions in G.lucidum.After a series of phenotypic and genetic analysis research,we found that the genes owned other functions in G.lucidum,being involved in regulation of the redox balance,cell wall synthesis,triterpenes biosynthesis and intracellular calcium signal,etc.To sum up,in this study,we highlight the roles of AP1 in mycelium growth,secondary metabolism,maintain the REDOX balance.We found the AP1 plays an important in the interaction of ROS signal and calcium signal,maintainning the balance of calcium in the body,affectting the growth of hypha.In addition,we also studied the REDOX system in G.lucidum by exploring the antioxidant mechanism of H2. |
