The Cervus nippon is considered a rare animal,and its products are used in nutritional ingredients,biological properties,and food safety,and have high utilization value.The C.nippon resources are abundant in China,such as Northeast Asian species,etc.At present,most of the C.nippon products are health products prepared from antlers as raw materials,and by-products such as the skin of C.nippon are underutilized.Most the skin of C.nippon are processed into leather,and a large number of high-quality collagen peptide resources are not if it is fully utilized,the breeder suffers serious economic losses.In this studies,the small molecule collagen peptide was prepared from the skin of C.nippon as raw material,and its physical and chemical properties and biological activities were studied,including antioxidant and melanin inhibition,which proved the high quality physical and chemical properties,antioxidant activity and melanin inhibition activity of the skin of C.nippon collagen peptide.In this studies,the optimal extraction process of collagen was determined by a single factor experiment and response surface optimization experiment,and finally determined the skin of C.nippon solid-liquid ratio of 1:40,p H 7.5,amount of compound protease added of 1600 U/g,51?,extraction for 5.5 h,and the best extraction rate was 75.11%.The molecular weight of the obtained collagen is less than 100 k Da,the hygienic index and sensory index meet the national standard,the solubility and emulsifying activity are the worst at p H 8,the water holding capacity finally reaches 83.30%within the measurement time.The optimal enzymatic hydrolysis conditions were established in this studies:the skin of C.nippon collagen peptide mass concentration of 2 mg/ml,p H 9,amount of compound protease added of 9800 U/g,50?,enzymatic hydrolysis for 3 h,and the final hydrolysis degree was 88.01%.The obtained collagen peptide is 6 k Da,the hygiene of the collagen peptide meets the national standard,the sensory quality is acceptable,and the physical and chemical properties include solubility,water absorption,water holding capacity,oil absorption,emulsifying activity,and emulsifying stability are better than that of collagen.It is shown that collagen peptides have better application properties than collagen.The biological activities of the skin of C.nippon collagen and collagen peptides were determined.With the increase of concentration,the antioxidant activity and the ability to inhibit melanin gradually increased.Collagen peptides also showed stronger biological activity than collagen.The ability to scavenge DPPH·,HO?,O2?-was50.01%,97.75%,50.38%,the reducing power was 0.365(700 nm),the inhibition rate of tyrosine mycophenolate was 60.73%,and the inhibition rate of L-dopa was 97.25%.In vitro gastrointestinal simulated digestion of collagen peptides was carried out.The results showed that after gastrointestinal digestion,the antioxidant activity and the ability to inhibit melanin were improved to a certain extent,which proved that collagen peptide still had a high degree of biological activity after entering the body.In conclusion,the preparation of the skin of C.nippon collagen peptides by enzymatic hydrolysis is a feasible process,which can provide technical support for the production of high value-added the skin of C.nippon health care products,provide data support for the application of the skin of C.nippon collagen peptides in functional food fields,and promote the sustainable development of the C.nippon industry development has important practical significance.
