Transforming growth-interacting factor-1(TGIF1)is a transcriptional repressor that regulates retinoic acid(RA)and transforming growth factor?(TGF-?)signaling pathways.Participate in mouse embryonic development,bone marrow differentiation and lipid metabolism.In humans,mutations in TGIF1 may lead to the occurrence of Holoprosencephaly(HPE)in human genetic diseases.TGIF1 mutations can also promote the development and metastasis of various malignant tumors through different pathways.Studies have shown that TGF-?signaling regulated by TGIF1 can regulate the differentiation and function of T cells and B cells,and TGIF1 is highly expressed in lymphocytes,but its function in the immune system is still unknown.Here,we explored the effect of Tgif1 gene knockout on mouse immune cell function.In this study,by using CRISPR/Cas9 system and microinjection technology,we constructed a Tgif1 knockout mouse model base on C57BL/6J background.Flow cytometry was used to analyze the differences of T and B cells between Tgif1 knockout mice and wild-type mice.We found that compared with wild-type mice,T and B cell subpopulations and natural killer cells from Tgif1-/-mouse immune organs had no significant differences.However,some of Tgif1-/-mice shows dysfunction on molars,resulting in a lean body.This may caused by the Tgif1 gene knockouts that lead to left-right asymmetry in mice and which has been confirmed by previous studies.Tgif1-/-mice with molars dysfunction showed significant changes in many lymphocyte subsets compared to wild-type mice,such as mature B cells,immature B cells,and T cells.However,the difference in molar-deficient mice may be due to the stress response caused by starvation,so we will not go into it.The Immunizations method will be used to further explore the effect of Tgif1 gene knockout on the differentiation and function of mouse-related immune cells.
