Membrane-reactive agents and cryopreservation of boar spermatozoa

embrane reactive agents (surfactants) may improve survival during cryopreservation of boar spermatozoa. This thesis investigated the effects of surfactants on post-thaw motility, acrosome integrity, intracellular ATP concentration, head plasma membrane (HPM) fluidity, intracellular calcium (Ca;Fluidity of unique domains in HPM isolated from fresh and cryopreserved boar spermatozoa was measured with fluorescent polarization using three fluorescent probes: cPNA, tPNA and DPH. Following cryopreservation in any extender, isolated HPM did not undergo the fluidity changes seen in the HPM from fresh spermatozoa. Egg yolk in the extender increased the fluidity of the DPH domain; presence of OEP but not pluronic prevented this.
