Objective: Alzheimer’s disease(AD)is a degenerative disease of the nerve system,which is called "dementia" in traditional Chinese medicine and has a complicated pathogenesis.In this study,we treat the 5×FAD mice with various combinations of separate component of huangqi san,to search for components which could relieve cognitive and pathological dysfunctions as effective as the compound huangqi san,thus provide theoretical basis for clinical prevention and treatment of AD.Methods:(1)Sixty 2-month old 5×FAD male mice were randomly divided into 5 groups using SPSS 22.0 software.Twelve wild type mice were used as control group.At 2 months of age,the drugs were administered separately for 3 months.(2)Behavioral experiments were carried out at the age of 5 months.Brain tissue sections were taken for IHC to observe the morphological and quantitative changes of neurons,astrocytes and microglia in the hippocampus and cortex of mice,and Thio-s staining was used to count the deposition of Aβ.Results:(1)3 months after the intervention of huangqi san and its separate components,there were no significant differences in the speed and distance of free movement in the open field and the proportion of time in the surrounding area among the mice in each group.(2)In the new object recognition experiment,the differentiation index of AD model group was significantly lower than that of the wild control group at 1H,and that of huangqi san group was significantly higher than that of AD model group.There was no significant difference between huangqi san group and huangqi + gegen group.(3)In the water maze experiment,the escape latency of mice in each group showed a general downward trend.The treatment group of huangqi san could significantly improve the duration and frequency of the AD mice in the target quadrant.There was no significant difference between huangqi san group and huangqi + gegen group.(4)Thio-s staining showed that the Aβ in the hippocampal and cortical areas of the mice in each treatment group were significantly lower than those in the AD group,and there was no significant difference between huangqi san and huangqi + gegen and huangqi + sangbaipi group.(5)Immunohistochemical results suggested that huangqi san could inhibit the activation and aggregation of microglias and astrocytes,and there was no significant difference between huangqi san and huangqi + gegen group and huangqi + sangbaipi group.Conclusion: Huangqi san could relieve the learning memory and spatial cognitive impairment of 5×FAD mice as well as AD-like pathology.Among the seperate parts,huangqi + gegen group has the best prevention and treatment effect on AD,close to the whole prescription of huangqi san,suggesting that huangqi and gegen are the main components of huangqi san in AD,which could take place to the original prescription.
