Objective:In this study,Enzyme-linked immunosorbent assay was used to detect serum platelet-derived growth factor-BB(PDGF-BB),platelet-derived growth factor receptor-β(PDGFR-β),Monocyte chemotactic factor-1(MCP-1),matrix metalloproteinase-9(MMP-9)expression level in patients with coronary heart disease and blood stasis,using RT-PCR technology to detect PDGFR-β mRNA expression level in peripheral venous blood mononuclear cells of patients with coronary heart disease and blood stasis syndrome.And by analyzing and comparing the differences in the expression levels of the above indicators between patients with coronary heart disease and healthy people,and then exploring the correlation between coronary heart disease and the above indicators.At the same time,analyze and compare the difference in expression level of the above indexes between patients with coronary heart disease and blood stasis syndrome and non-blood stasis syndrome,and then explore the correlation between coronary heart disease blood stasis syndrome and appeal index.Provide more biological basis and reference basis for the pathological mechanism and clinical diagnosis of coronary heart disease and blood stasis.Methods:From September 2019 to January 2020,128 patients with coronary heart disease who were hospitalized in the Department of Cardiology,Guang’anmen Hospital,Chinese Academy of Chinese Medical Sciences were selected,and included 30 healthy examinees in the Southern District Medical Examination Center of Guang’anmen Hospital as healthy.Control group.According to the "Diagnostic Criteria for Blood Stasis Syndrome of Coronary Heart Disease" formulated by the Professional Committee for Promoting Blood Circulation and Stasis of the Chinese Society of Integrated Chinese and Western Medicine in 2016,128 patients with coronary heart disease were divided into blood stasis syndrome group(87 cases)and non-blood stasis syndrome group(41 cases).On the morning of the second day of admission,the fasting venous blood of all subjects was drawn,and the levels of PDGF-BB,PDGFR-β,MCP-1,and MMP-9 in peripheral venous blood were detected by enzyme-linked immunosorbent assay(ELISA),and RT-PCR technology was used to detect the expression level of PDGFR-β mRNA in peripheral venous blood mononuclear cells,and at the same time collect and organize the general clinical data,blood routine,biochemical and other relevant clinical test data of all enrolled subjects.The data obtained was statistically analyzed and processed using SPSS 22.0 statistical software.Result:1.The serum levels of PDGF-BB,PDGFR-β,MCP-1 and MMP-9 in the coronary heart disease group were significantly higher than those in the healthy control group,and the difference was statistically significant(p<0.01).2.The expression level of PDGFR-β mRNA in peripheral blood mononuclear cells of the coronary heart disease group was significantly higher than that in the healthy control group,and the difference was statistically significant(p<0.01).3.Coronary heart disease is positively correlated with PDGF-BB,PDGFR-β,MCP-1,MMP-9,and PDGFR-β mRNA.4.The levels of serum PDGF-BB,PDGFR-β,MCP-1 and MMP-9 in the coronary heart disease blood stasis syndrome group were significantly higher than those in the coronary heart disease non-blood stasis syndrome group,and the difference was statistically significant(p<0.01).5.The expression level of PDGFR-β mRNA in peripheral blood mononuclear cells of coronary heart disease blood stasis syndrome group was significantly higher than that of coronary heart disease non-blood stasis syndrome group,the difference was statistically significant(p<0.01).6.Coronary heart disease blood stasis syndrome was positively correlated with PDGF-BB,PDGFR-β,MCP-1,MMP-9,and PDGFR-β mRNA.Conclusion:1.Serum PDGF-BB,PDGFR-β,MCP-1 and MMP-9 levels were elevated in patients with coronary heart disease.2.PDGFR-β mRNA expression in peripheral blood mononuclear cells of patients with coronary heart disease increased.3.Coronary heart disease is positively correlated with PDGF-BB,PDGFR-β,MCP-1,MMP-9,and PDGFR-β mRNA.4.The levels of PDGF-BB,PDGFR-β,MCP-1 and MMP-9 in patients with coronary heart disease and blood stasis syndrome increased.5.PDGFR-β mRNA expression in peripheral blood mononuclear cells of patients with coronary heart disease and blood stasis syndrome increased.6.Coronary heart disease blood stasis syndrome was positively correlated with PDGF-BB,PDGFR-β,MCP-1,MMP-9,and PDGFR-β mRNA.
