Purpose: To study the regulation of Kv1.4 and Kv4.2 on the expression of transient outward potassium channel related proteins in myocardial cells of rats with acute myocardial infarction induced by high-fat feeding and isoproterenol,and to explore the possible mechanism of the improvement of acute myocardial infarction by Acupuncture and drug combination at the level of ion channel,and to verify its advantages.Material and method:sixty five healthy male SD rats were randomly selected as the control group,and saline was injected subcutaneously into the inner root of the extremities twice,with an interval of 24 hours.In addition to the control group,the other rats were fed with normal diet to induce hypercholesterolemia for 3 weeks.The acute myocardial infarction model was also made by injecting isoproterenol(85mg / kg)subcutaneously at the root of four limbs.Compared with the ECG changes before and after the model establishment,40 rats were randomly divided into model group,acupuncture combined with medicine group,acupuncture group and Western medicine group,10 rats in each group.The blank group and the model group were administrated with normal saline once a day in the morning and evening;the combination group was administrated with Xinjing Tongmai Decoction once a day in the morning and evening,and Neiguan point,Xinshu point and Zusanli point were needled once a day;the acupuncture group was administrated with Neiguan point,Xinshu point and Zusanli point once a day;the western medicine group was administrated with aspirin and isosorbide mononitrate solution once a day in the morning and evening.Huatuo brand acupuncture(0.18 mm * 25MM)was used to prick the corresponding acupoints of rats subcutaneously for about 2mm.After the rats were stable,density wave was used with a frequency of 2-20 hz and a strength of 2m A.Acupuncture treatment was carried out for 20 minutes each time.In the course of treatment,the survival status of rats,the changes of ECG ST segment before and after treatment were observed,and the expression of protein related to outward potassium channel(Kv1.4,Kv4.2)was detected by Western blot.Results:1.Compared with the control group,the ST wave of the acupuncture combined with medicine group,the acupuncture group and the western medicine group decreased(P < 0.05),among which the ECG changes of the acupuncture combined with medicine group were the most obvious,but there was no significant difference between the acupuncture group and the western medicine group(P > 0.05).2.Compared with the control group,the protein expression of Kv1.4 and Kv4.2 in the model group was significantly decreased(P < 0.05).After 15 days of treatment,compared with the model group,it was found that the expression of index protein in the acupuncture drug combination group,the acupuncture group and the western medicine group were all increased(P < 0.05),and the increase of index protein in the acupuncture drug combination group was significantly better than that in the acupuncture group and the western medicine group(P <0.05);there was no significant difference in protein expression between the acupuncture group and the western medicine group(P > 0.05))3.Compared with the control group,the expression of CTRP3 protein in the serum of the model group decreased significantly(P < 0.05).After the intervention treatment,compared with the model group,it was found that the protein expression of the indexes in the acupuncture drug combination group,the acupuncture group and the western medicine group was significantly increased,the difference was statistically significant(P > 0.05).3.Compared with the control group,the expression of CTRP3 protein in the serum of the model group decreased significantly(P < 0.05).After the intervention treatment,compared with the model group,the protein expression of each index in the acupuncture drug combination group and the acupuncture group increased significantly(P < 0.05).2.Conclusion:1.When myocardial infarction occurs in rats,the ST segment of ECG will show elevated changes.The abnormal changes of ST segment can be effectively improved by the treatment of qijingtongmai decoction combined with acupuncture at corresponding points.In addition,the decrease range of acupuncture combined with medicine group was larger than that of other groups.2.The expression of Kv1.4 and Kv4.2 in normal rat cardiomyocytes was normal.When myocardial infarction occurred,the expression of Kv1.4 and Kv4.2 decreased significantly.The expression of related proteins can be increased after the treatment of qijingtongmai decoction combined with acupuncture at corresponding acupoints.3.In the acupuncture group,the expression of transient outward potassium channel related proteins in cardiomyocytes was regulated,while the data showed that the regulation range in the acupuncture drug combination group was greater.4.During myocardial infarction,the content of ctrp3 in serum was lower than that in normal rats.The combination of acupuncture and medicine can effectively increase its expression,and the increase of the combination of acupuncture and medicine is larger than that of other groups,which proves that the combination of acupuncture and medicine is more effective in the treatment of acute myocardial infarction.
