| Purpose:To explore the possibility of inducing apoptosis of gastric cancer cell SGC-7901 by modified xiangsha liujunzi decoction,and To elucidate its mechanism,So as to provide a laboratory basis for clinical application of modified xiangsha liujunzi decoction in the treatment of gastric cancer.Method:In this paper,using in vitro culture technology,the modified xiangsha liujunzi decoction,was divided into low,medium and high groups according to the doses of 1.55g/kg,3.1g/kg and6.3g/kg.The 5-Fu of 50umol/l was set as the 5-Fu observation group,and a blank control group was set up.Samples were collected after the above reagents were applied to gastric cancer cell line SGC-7901 at logarithmic growth stage for 48 h.The apoptotic cycle of gastric cancer cell line SGC-7901 was detected by flow cytometry analysis,and the same sample was repeated for three times.Immunocytochemistry was used to detect the expression levels of proteins in gastric cancer cell lines SGC-7901 Bax,Bcl-2 and Caspase-3 treated with modified xiangsha liujunzi decoction.The same samples were repeated for three times.SPSS23.0 software was used to analyze the data.The cell percentage and expression levels of Bax,bcl-2 and Caspase-3 in cells of different cell cycles in each group were expressed as (?)±S,and the mean values of samples were compared by one-way anova.Results:1.G1 phase ratios of blank control group,low-dose TCM group,medium-dose TCM group,high-dose TCM group and 5-fu group were as follows:89.34±1.42,87.80±1.93,84.06±2.52,82.87±1.71,82.11±2.76。S phase ratios are respectively:8.36±0.65,7.75±0.80,7.46±1.55,6.31±1.51,6.11±2.09。G2 phase ratios are respectively:2.09±1.07,4.45±1.69,8.49±2.03,10.83±2.46,11.79±2.24。2.After Bcl-2 staining,the number of brown-yellow particles in the cells of the control group was significantly increased,and most of the cells were strongly positive.The positive expression rate of Bcl-2 protein decreased with the increase of dose and time,and the proportion of cells with strong positive staining decreased obviously.The number of brown-yellow granules in the cells of the control group after Bax staining was small,and most cells were weakly positive.The positive expression rates of Bax and Caspase-3 protein increased with the increase of dose and time,and the proportion of cells with strong positive staining increased significantly..After Caspase-3 staining,the number of brown-yellow particles in the cells of the control group was small,and most cells were weakly positive.After the treatment of SGC-7901 cells with different concentrations of flavor-added xiangsha-liujunzi decoction,the positive expression rate of Caspase-3 protein increased with the increase of dose and time,and the proportion of cells with strong positive staining increased significantly.3.The expression of Bcl-2 protein in the low concentration group was lower than that in the medium concentration group and the high concentration group(p<0.05).Bax protein expression in high concentration group was higher than that in low concentration group and medium concentration group(p<0.05).The expression of Caspase-3 protein in the high-concentration group was higher than that in the low-concentration group and the medium-concentration group(p<0.05).Conclusion:1.Modified Xiangsha Liujunzi Decoction can block SGC-7901 cells in G2 phase and induce their apoptosis.2.The protein expression of SGC-7901 was observed after the effect of Modified Xiangsha Liujunzi Decoction at different concentrations.The positive expression rate of Bcl-2decreased with the increase of dose and time,while the positive expression rate of Bax and Caspase-3 increased with the increase of dose and time.3.The effect of high concentration of Modified Xiangsha Liujunzi Decoctionon the cellular protein Bax and Caspase-3 of gastric cancer SGC-7901 was significantly better than that of low and medium concentrations.However,the effect of low-concentration Modified Xiangsha Liujunzi Decoctionon on the cellular protein Bcl-2 of gastric cancer SGC-7901 was significantly better than that of medium-high concentration.3.The effect of high concentration of Modified Xiangsha Liujunzi Decoctionon on apoptosis of gastric cancer SGC-7901 was significantly better than that of low and medium concentration.4.The expression of Bcl-2 was down-regulated and the expression of Bax and Caspase-3 was up-regulated,which may be related to apoptosis. |
