Based On The JAK/STAT Signal Pathway To Study The Protective Mechanism Of Warm Needling Moxibustion On Blood Vessels In Spontaneously Hypertensive Rats

Purpose:Under the guidance of the traditional Chinese medicine theory.Acupuncture and warming moxibustion have clear and definite effects in the aspects to occur and develop the hypertension and to delay effectively target organic damage and to lower relevant complications.This experiment uses spontaneously hypertensive rats as models and uses isogeny ortho-arteriotony rats as matched group.They will be inflicted needle warming moxibustion,acupuncture and moxibustion on the hole of zusanli point.then we will observe the influence to pressure and morphology of aorta abdominallis of spontaneously hypertensive rats.With WB and q PCR etc technical detections,we will detect the expressions of coherent signals in JAK/STAT passageway and that is aimed at discovering the mechanism of action that acupuncture protects arterial vascular function in the body of hypertension and then protect target organs with damage for providing experiment gists in prophylaxis and treatment hypertension by acupuncture.Material and method: Ten Consanguinity WKY rats are used only as matched group,Fourty consanguinity spontaneously hypertensive rats are divided to model group,needle warming moxibustion group,acupuncture group and moxibustion group according to random number table method.After buying the rats,they are measured unitively blood pressure and are feeded conventional fodder and drinked water by itself for a week in order to adapting experimental environment.Later turning into official experimental stage,matched group and model group are only fixed everyday without any treatment.Needle warming moxibustion group is needled by filiform on both sides of Zuzanli points,depth of 5 mm,and in the meantime inflicted moxibustion on the top of needle handle.Acupuncture group is needled by filiform on both sides of Zuzanli points,depth of 5 mm.Moxibustion group is inflicted moxa-moxibustion alone.These operations are proceeded on nine o’clock at one time everyday,holding 20 minutes each time,continueing 15 days.The rats are monitored blood pressure at the same time on the day of prior treatment and on the day of 1,3,5,7,9,11,13,15,17,19and21 of post treatment.After the treatment,waiting eight days,we take blood in the abdominal aorta after anesthesia of rats,get abdominal aorta tissue,detect pathologic morphological change of abdominal aorta blood vessel with HE dyeing observation,detect expression of related signal pathways with q PCR,WB methed.Results: 1.Needle warming moxibustion can specificly regulate JAK/STAT signal pathway.The experiment results show that needle warming moxibustion on Zusanli point can specificly lower STAT1 m RNA expression,can lower the degree of protein expression of STAT1,JAK1 and p-STAT1,which impress backward domino effect,lower blood pressure,with it can protect blood vessel and target orgen meanwhile.2.Needle warming moxibustion can improve pathomorphism of blood vessel.The experiment results show that needle warming moxibustion treatment can improve cell arrangement of arterial wall and the surface condition of inner membrance inordinately,can perfect the phenomenon of endothelial cells hyperplasia or cell fall off inordinately,can improve cell arrangement,hyperplasia,large of smooth muscle at tunica media and the interval and arrangement of elastic fibers,make inordinately the surface of perithelial cells to turn smooth without the phenomenon of hyperplasia and large.3.Needle warming moxibustion can reduce blood pressure of SHR rats.The experiment results show that all treatment groups come to lower systolic pressure and diastolic pressure of SHR rats inordinately.The lowering scope of systolic pressure in needle warming moxibustion group is about 45 mm Hg;The lowering scope of systolic pressure in acupuncture group is about 20 mm Hg;The lowering scope of systolic pressure in moxibustion group is about 35 mm Hg;and the onset time in proper order from fast to slow is needle warming moxibustion group,acupuncture group and moxibustion group.The lowering scope of diastolic ic pressure in needle warming moxibustion group is about 30 mm Hg;the lowering scope of diastolic ic pressure in acupunction group is about 15 mm Hg;The lowering scope of diastolic ic pressure in moxibustion group is about 25 mm Hg.As shown above,needle warming moxibusion treatment can specificly degree the systolic pressure and diastolic pressure of SHR rats.Meanwhile needle warming moxibustion express the fastest onset time.Thus it can be seen that the lowering scope of systolic pressure is greater than the lowering scope of diastolic pressure.In addition,the blood pressure in acupuncture group rise again afer therapy discontinued.Conclusion: 1.Needle warming moxibustion Zusanli point can protect vascular structure and function of SHR rats.Needle warming moxibustion Zusanli point can perfect blood vessel form and inflammatory response to guard blood vessel especially artery blood vessel for preventing target organic from damage and delaying the appearing of complications in hypertension disease.2.Needle warming moxibustion Zusanli point can play a role of protective effect to blood vessel by means of adjusting JAK/STAT relevant signal path.Needle warming moxibustion Zusanli point can constrain JAK/STAT signal path in blood vessel,thus go down expression level and protein level of STAT1,JAK1 and JAK2,restrain domino offect in backward position and lower blood pressure.This may be one of the normal operation mechanism to protect blood vessel in hypertension disease.
