Objective:The process of cordyceps militaris fermentation to gastrodia elata was optimized to investigate the anti-depression effect of cordyceps militaris and gastrodia elata fermentation products.Methods:The five factors of fermentation time,carbon source addition,nitrogen source addition,inorganic salt addition and Gastrodia filler were optimized.The content of cordycepin and gastrodin in the fermentation products of Cordyceps militaris-Gastrodia Bi-directional fermentation system was used as the detection index,and single factor experiment design was carried out.The Cordyceps militaris-Gastrodia fermentation product was produced in batches under the optimal conditions selected.CUMS model rats were intragastrically administered with high,medium and low doses of fermentation product aqueous extracts.During the experiment,the fur and morphological changes of the rats were observed.Before molding,after molding and after administration,the behavioral changes of rat body weight,Sucrosepreference test,Open field test and Morris water maze were detected.The expression level of GluA1 and the phosphorylation level of GluA1 Ser845 in the prefrontal cortex and hippocampus of rats were detected by western blot after behavioral examination.Results:①The optimal technology of cordyceps militaris-gastrodia elata bi-directional fermentation system is:fermentation time 12d,soluble starch 20g/L,peptone 1g/L,no inorganic salt added and 4%gastrodia elata packing amount.②Fermentation product can improve the symptoms of fur luster loss and poor behavior and posture of rats after stimulation of modeling;Fermentation products can improve the weight gain,sucrose water preference,horizontal movement distance and vertical activity times of the CUMS rats in the Open field test,and shorten the time of finding the platform in the Morris water maze of the depression model rats.③The results showed that The fermentation products had no significant effect on the expression level of GluA1 in the prefrontal cortex and hippocampus of rats;fermentation products could increase the phosphorylation level of GluAl Ser845 in the prefrontal cortex of rats.Similarly,fermentation products could increase the phosphorylation level of GluAl Ser845 in the hippocampus.Conclusion:①After the optimization of Cordyceps militaris-Gastrodia Bi-directional fermentation system,the content of cordycepin in the fermentation products increased.②In addition,the fermentation product has an anti-depression effect on the CUMS model rats,and its effect is better than that of Cordyceps militaris and Gastrodia.③The anti-depression mechanism of the water extract of f cordyceps militaris-Gastrodia fermentation product is related to its ability to increase the phosphorylation level of GluA1 Ser845 in the prefrontal cortex and hippocampus of rats.
