Objective: The effect of shennaofuyuan Decoction on HUC-MSCs)proliferation and the impact on its cycle,to explore the best time and dose-response relationship of shennaofuyuan Decoction and HUC MSCs in animal experiments,in order to accumulate experimental basis for the combination of the two in the intervention of ischemic stroke neuroprotection.Methods: 1.High,medium and low dose shennaofuyuan decoction was used to infuse SPF grade SD rats to prepare the serum containing medicine,and the normal serum was used as the control.2.By CCK-8,the effect of serum containing drugs on cell proliferation was observed.The serum concentration of 20% and the HUC MSCs used for subsequent intervention in vitro culture were selected.3.The cell cycle of each group at different times was detected by flow cytometry.4.The protein expression levels of β-Catenin,c-myc and cyclin D1 were detected by Western blot.Results: 1.CCK-8 method: 10% of the seru containing shennaofuyuan decoction had the best proliferative activity in 48 hours.2.The results of flow cytometry showed that 10% shennaofuyuan Decoction serum could induce the cell cycle to G2 / M phase 48 hours after treatment.3.Western blot detection: 10% shennaofuyuan containing serum can promote the expression of β-Catenin,c-myc,cyclin D1 protein to a certain extent,and it is the best in 48 hours.Conclusion: Shennaofuyuan decoction can promote the proliferation of HUC MSCs and induce the cell cycle to G2 / M phase,and the best time is in 48 hours with 10% serum.It can provide the basis for the combined treatment of ischemic stroke and protect neurons.This effect may be related to the expression of β-Catenin,c-myc and Cyclin D1. |
