| Arctium lappa is a biennial herb,Dicotyledoneac,Asteraceae,and Arctium,It is widely distributed in China,Western Europe,Russia,North America,South America and other regions.It enjoys a warm and humid climate,is cold and heat resistant and is mostly found in valleys,hillsides,forest margins and forests,with an altitude of 750 to 3500 metres.Its roots can be used as a traditional medicinal plant or as a vegetable.Fructus Arctii is a dry and ripe fruit of burdock,which is widely used as a traditional medicine for sweating agents,diuretics and blood purifying agents.It has the effects of evacuating wind and heat,swelling and detoxification.It is clinically used for the treatment of wind-heat cough,sore throat,sore swollen poison[1].The compounds currently isolated from such plants are lignans,terpenoids,phenolic acids,flavonoids and other components[2].This dissertation is mainly composed of four chapters.The isolation and identification of compounds from Fructus Arctii was described in Chapter 1.The volatile oil components in different provinces burdock was analyzed in chapter 2.The fingerprint chromatogram of Arctium lappa L.from different cultivation places by HPLC was described in Chapter 3.The fourth chapter described the research on the main types of compounds and pharmacological actions in the burdock.[Objective]Analysis of the chemical constituents and volatile oil components of Yunnan burdock.[Methods]The first chapter used the chromatographic technique to study the chemical constituents of Yunnan burdock.The burdock was extracted by using 90%methanol-water,and the crude extract was separated and purified.The separation and purification were mainly carried out by silica gel column chromatography,Sephadex LH-20,MCI GEL,and RP-18 silica gel on repeated column chromatography,and finally purified by semi-preparative RP-HPLC.The structure of chemical components was identified based on the physicochemical properties of the compound and the spectral analysis data.Compounds 1*-3*were tested for their anti-TMV activity.The anti-TMV activity was tested using the half-leaf method.Ningnanmycin with inhibition rate of 31.2%,was used as a positive control.The anti-MRSA agar disk diffusion assay was used to measure the activity of methicillin-resistant Staphylococcus aureus(anti-MRSA),2’,7’-dichlorofluorescein diacetate(DCFH)method was used to detect oxidation products to determine antioxidant activity.In the second chapter,the volatile oil components of Fructus Arctii from six six different areas were analyzed by simultaneous distillation extraction and gas chromatography-mass spectrometry.In the third chapter,the fingerprint of Arctium lappa L.from different origins was studied by high performance liquid chromatography.[Results]Twenty-three compounds were isolated from Yunnan burdock,including five new compounds(1*-5*)and 18 known conpounds(6-23).The results revealed that compounds 1*,2*and 3*showed high anti-TMV activity with inhibition rates of 33.5%,32.8%,and 34.2%,at the concentration of 20 μM,respectively.These rates were higher than that of positive control.The inhibition of the activity of the new compound 4*and 5*against methicillin-resistant Staphylococcus aureus(MRSA)was weak,and the diameter of the inhibition zone(IZD)was 10.8±1.0 and 9.3±0.8 mm,respectively.The antioxidant activities of Compounds 4*and 5*were also tested and showed good antioxidant activity with IC50 values of 3.85 and 3.47 μg/mL,respectively.The analysis of volatile oil of burdock from six different areas showed that there were some differences in the composition of volatile oil from different areas.The fingerprint chromatogram of burdock from nine different origins showed that the similarity was greater than 0.99,which indicated that the quality of burdock from different origins had good homogeneity. |
