Part Ⅰ.THE EXPRESSION OF HOTAIR,COLⅡANDMMP3 IN HUMAN NORMALAND DEGENERATEDNUCLEUS PULPOSUS AbstractObjective: To investigate the expression of HOTAIR,Col Ⅱ and MMP3 in human normal and degenerated nucleus pulposus.Methods: Six patients with lumbar burst fractures or scoliosis were randomly selected as the normal control group(NC group),and 24 patients with degenerative disc disease(Under Pfirrmann classification,6in the grade Ⅲ,10 in the grade Ⅳ,and 8 in the grade Ⅴ)were used as the experimental group(IDD group).The q RT-PCR was used to detect the expression of HOTAIR,Col Ⅱ and MMP3 both in normal and degenerated nucleus pulposus.Results: The expression level of HOTAIR and Col II in the normal nucleus pulposus tissue were higher than those in degenerated group,and the expression level in the degenerated group decreased with increasing grade(P<0.01).The expression level of MMP-3 in the normal nucleus pulposus tissue was lower than those in the degenerated group,and the expression level in the degenerated group increased with increasing degenerated grade(P<0.01).Conclusion: The degenerated degree of human nucleus pulposuswas negatively correlated with the expression level of HOTAIR and Col II,and positively correlated with the level of MMP3 expression.Part Ⅱ.TO STUDY THE CORRELATION BETWEENHOTAIR AND COLⅡWITH MMP3 EXPRESSION INHUMAN NUCLEUS PULPOSUS CELLS AbstractObjective: To explore the correlation between HOTAIR and the expression of Col Ⅱ and MMP3 in human nucleus pulposus cells.Methods: HOTAIR knockout group(si HOTAIR-1,si HOTAIR-2,si HOTAIR-3)and HOTAIR overexpression group(Vector-HOTAIR)were constructed.HOTAIR knockout group and overexpression group were transfected into nucleus pulposus cells,and q RT-PCR and western blot were used to detect the expression levels of Col Ⅱ and MMP3.Results: In the HOTAIR knockout group,the expression level of ColⅡdecreased,while the expression level of MMP3 increased.In the HOTAIR overexpression group,the expression level of Col Ⅱ increased and the expression level of MMP3 decreased.Conclusion: HOTAIR is correlated with Col II and MMP3 expression,which can up-regulating Col II expression and down-regulating MMP3 expression.
