Objective:Through rigorous scientific research design and randomized controlled experiments,based on the Nrf2/HO-1 signaling pathway to explore the effect of yam-rhizoma imperatae chinense on the contrast agent nephropathy(CIN)oxidative stress in rats Experimental basis.Methods:The 48 healthy and clean male SD rats were randomly divided into a blank group,a model group,a yam-rhizoma imperatae group[6.3g/(kg·d)],a yam group[3.15g/(kg·d)],and a rhizoma imperatae group[3.15g/(kg·d)] and N-acetylcysteine(NAC)control group [252mg/(kg·d)],8animals in each group.The medicine pair group,yam group,and rhizoma imperatae group were given the corresponding drug decoction according to the dose 7d before the model building,the NAC group was given the corresponding drug aqueous solution according to the dose,the blank group and the model group were given the same volume of distilled water,daily Once a day,the drug is given for 7 consecutive days before modeling.After the corresponding treatments were given to each group,the model group,drug pair group,yam group,rhizoma imperatae group and NAC group prepared CIN models.Blood was collected from the abdominal aorta 24 hours after the model was made to detect the serum creatinine(Scr)and urea nitrogen(BUN)levels of the rats;kidney tissues were taken for hematoxylin-eosin(HE)staining to observe the pathological and morphological changes of the kidney tissues;immune group The expression of nuclear factor E2 related factor(Nrf2)and heme oxygenase-1(HO-1)in renal tissue was detected by chemical staining.Results:1.Blood biochemical index detection After 24 hours of modeling,the levels of serum Scr and BUN in the model group were significantly higher than those in the normal group,and the difference was statistically significant(P<0.05);the levels of serum Scr and BUN in the drug-pair group and NAC control group and the modelCompared with the normal group,the difference was statistically significant(P<0.05);there was no significant difference in serum Scr and BUN between the drug-pair group and the NAC control group(P>0.05).Serum Scr and BUN in the yam group and rhizoma imperatae group were lower than the model group,but the difference was not statistically significant(P>0.05).2.HE staining to observe the histopathological changes of the kidneys in each group There were no obvious lesions in the glomeruli,renal tubules and renal interstitium in the normal group.In the model group,the renal tubular epithelial cells were extensively vacuolar-like degeneration,some of the renal tubular necrosis,the brush border of the epithelial cells fell off,the renal tubular structure was destroyed,and there were erythrocyte cast and protein cast,edema and inflammatory cell infiltration.Compared with the model group,the drug-pair group and the NAC group were significantly improved.The vacuole-like area of renal tubular epithelial cells was significantly reduced.The structure was relatively complete.The renal tubular epithelial cells were slightly swollen.The yam group and rhizoma imperatae group showed no significant improvement compared with the model group.3.Observe the expression of Nrf2 and HO-1 in kidney tissueCompared with the normal group,the expression of Nrf2 and HO-1 in the renal tissue of the model group increased,and the difference was statistically significant(P<0.05);the expression and model of Nrf2 and HO-1 in the renal tissue of the drug-pair group and the NAC group The difference was statistically significant between the two groups and the difference was statistically significant(P<0.05);there was no statistically significant difference in the expression of Nrf2 and HO-1 protein in the renal tissue between the drug pair group and the NAC group(P>0.05);The expression of Nrf2 and HO-1 in the kidney tissue of the group was not statistically different from that of the model group(P>0.05).Conclusion:1.Chinese yam-white root "drug pair" can reduce serum creatinine and urea nitrogen levels in rats with contrast nephropathy,and improve pathological damage of contrast agent to rat kidney.2.Nrf2/HO-l pathway is involved in the pathogenesis of CIN.yam-whit root "drug pair" can increase the expression of Nrf2、HO-l protein,improve the antioxidant stress function of rats to protect the kidney and prevent the acute kidney injury caused by contrast agent.
