| Objective:Shenshan solid beverage is a kind of medicinal food drink and is composed of water extracts of eight kinds of herbs.The main function of the product is to enhance the function of spleen and kidney.Because some of herbs use in the product have antitumor activity,therefore,the purpose of this study is to explore the potential clinical value of Shenshan solid beverage by studying the effect of Shenshan solid beverage on human colorectal cancer cell line HT-29 in vitro.Methods:1.HT-29 cells cultured in 96-well plate were treated with complete medium containing different concentrations of Shenshan solid Bevera ge(0,2.5,5.0,10.0,20.0 mg/ml)for 48 hours,and cell growth was measured by using CCK-8 assay.2.The HT-29 cells were cultured with different concentrations of Shenshan solid beverage(0,5.0,10.0 mg/ml).The morphology of the cultured cells was observed under microscope.3.Hoechst 33258 staining method was used to detect apoptosis after the intervention of Shenshan solid beverage.4.The migration ability of HT-29 cells was monitored by Transwell assay after the intervention of Shenshan solid Beve rage.Results:1.Shenshan solid beverage inhibited HT-29 cell growth with dose-dependent effect(P <0.05).Its IC50 value is 8.96 mg/ml.According to its IC50 value,5.0 mg/ml and 10.0 mg/ml Shenshan solid beverage were selected as the high and low concentration of later experiments.2.Under microscope,the HT-29 cells become round,smaller,and fell off after Shenshan solid beverage treatment for 24 hours.3.Hoechst 33258 staining showed that the Shenshan solid beverage induced HT-29 cell apoptosis.4.Transwell assay showed that the number of HT-29 cells reaching the bottom of Transwell chamber in Shenshan solid Beverage group was significantly reduced,and Shenshan solid Beverage inhibited the migration ability of HT-29 cells.Conclusions:Beside its function in modulating spleen and kidney,this preliminary study suggested that the Shenshan solid beverage inhibited the proliferation and migration of human colorectal cancer HT-29 cells.One possible cause is the induction of cellular apoptosis. |
