Objective: To screen the optimal anti-proliferative drugs against human breast cancer cell lines MCF-7,MDA-MB-231 and SK-BR-3 from the total extracts of Stellera chamaejasme and their monomers,and observe the three The effects of human breast cancer cell cycle and apoptosis,and preliminary study of its mechanism.Materials and methods: 1.Subjects: Human breast cancer cell lines MCF-7,MDA-MB-231,SK-BR-3,and logarithmic growth phase cells were used for experiments.2.Experimental drugs: total extract of Stellera chamaejasme,Chamaejasmenin,Chamaejasmenin A,Chamaejasmenin C.3.Method: The effect of total extract of Stellera chamaejasme L.on the proliferation of three cell lines was detected by CCK-8 method.The drug concentrations were 10mg/ml,50mg/ml,100mg/ml,300mg/ml,500mg/ml,1000mg./ml.The effects of Chamaejasmenin,Chamaejasmenin A and Chamaejasmenin C on the proliferation of three cell lines were detected by CCK-8 method.;The effect of Chamaejasmenin A on apoptosis and cell cycle of three cell lines was detected by Western blotting.The expression of FAS,FASL and Caspase-3 protein in MDA-MB-231 cells treated with Chamaejasmenin A for 24 h was determined by Western Blot.Mechanisms that promote apoptosis in breast cancer cells.Results: 1.The extract of Stellera chamaejasme L.inhibited the proliferation of three cell lines.The IC50 of MCF-7,MDA-MBA-231 and SK-BR-3 were 286.76mg/ml and 486.69mg/ml,respectively.754.79mg/ml;and the cell cycle of the three cell lines were arrested in the S phase.2.Chamaejasmenin,Chamaejasmenin A and Chamaejasmenin C inhibited the proliferation of three breast cancer cell lines,and IC50 of wolf toxin on three cell lines MCF-7,MDA-MB-231 and SK-BR-3 75.81mg/ml,80.42mg/ml and 107.30mg/ml,respectively;the IC50 of Chamaejasmenin A on MCF-7 cells,MDA-MB-231 cells and SK-BR-3 cells were32.99mg/ml and 62.49mg/ml,respectively.And 68.96mg/ml;the IC50 of Chamaejasmenin Cto MCF-7,MDA-MB-231,and SK-BR-3 cell lines were 47.18mg/ml,98.64 mg/ml,and 94.31mg/ml,respectively.3.Chamaejasmenin A can arrest the cell cycle of all three cell lines in G1 phase and induce apoptosis of three cell lines.After treatment with Chamaejasmenin A for 24 h,the expression of FAS/FASL and Caspase-3 protein in MDA-MB-231 cells were up-regulated.Conclusion: 1.The proliferation of the three cell lines of breast cancer MCF-7,MDA-MB-231 and SK-BR-3 by the total extract of Stellera chamaejasme L.and its monomeric compounds,Chamaejasmenin A,Chamaejasmenin and Chamaejasmenin C All of them have inhibitory effects,in which Chamaejasmenin A has the highest inhibition rate on cell proliferation,the inhibition rate is the highest in MCF-7 cells,followed by MDA-MB-231 and SK-BR-3.2.The total extract of Stellera chamaejasme can block the three breast cancer cell lines in the S phase;the Chamaejasmenin A can block the three breast cancer cell lines in the G1 phase and promote the three cell lines.Apoptosis,apoptosis rate MDA-MB-231 highest,MCF-7followed by SK-BR-3.3.Chamaejasmenin A acts on MDA-MB-231 cell line,which can up-regulate the expression of FAS,FASL and Caspase-3 proteins,indicating that Chamaejasmenin A promotes apoptosis of MDA-MB-231 cells by expressing FAS and FASL.Up-regulated to achieve.
