Purpose:On the basis of pharmacodynamics,acute toxicity and clinical efficacy of KOA patients,the differential cytokines in blood samples of KOA patients were analyzed by antibody protein chip technique.To seek the changes of human blood samples related cytokines in the course of treating KOA with Xitong Kang preparation,and to provide the basis for further study on the mechanism of action.Material and method:Serum samples of 4 patients with KOA before and after taking Xitongkang were selected.Cytokine analysis was performed on human serum samples using Ray Bio ?antibody protein microarray.A self-control method was used to screen the different cytokines in the serum samples of patients before and after treatment.Results:1.Four different cytokines were detected in serum samples before and after treatment with 40 kinds of antibody protein microarray in four subjects taking Xitongkang.There are 2 kinds of up-regulated cytokines and 2 kinds of down-regulated cytokines.2.The levels of SIL-6R and TNF-α in serum of patients with KOA were down-regulated by Xitongkang preparation(P < 0.05).The serum levels of PDGF-BB and IFN-γcould be up-regulated in patients with KOA,but there was no significant difference(P > 0.05).Conclusion:1.Xitongkang can decrease the level of serum SIL-6R,TNF-α cytokines and increase the level of PDGF-BB cytokines in patients with KOA(P<0.05).2.The effect of Xitong Kang on human body can regulate some related cytokines in the serum of KOA patients.It is proved that the effectiveness of Xitong Kang is worth popularizing and applying in clinical practice.3.The experiment proved that the application of antibody protein chip technology in the study of Chinese patent drug therapy KOA screening related protein markers experiment effect is obvious,and through antibody protein chip technology to find and screen serum protein markers operation is simple,worth popularizing.. |
