Purpose:To establish a rat model of type 2 diabetes(T2DM)by diet-induced combination therapy,and to observe the PTP1 B pathway in rat liver tissue against the background of insulin resistance(IR)confirmed by overexpression of PTP1 B.The expression of IRS-2 and downstream PI3 K,thus clarifying the changes in the levels of protein and mRNA of the indicators after intervention with traditional Chinese medicine compound Yitangkang.It is proposed to prove the mechanism of action of Chinese herbal compound in the treatment of T2 DM to improve liver IR and the target of Chinese medicine.Material and method:50 SPF grade Wistar male rats were purchased and weighed(220 ± 20)g.Adapted feeding for 1 week,no adverse reactions were included in the formal experiment.Rats were numbered according to body weight,and then divided into blank groups according to the random number table method,and fed to normal diet.The remaining 42 were fed with high fat diet.After 4 weeks,rats were injected intraperitoneally with STZ at a dose of 45 mg/kg body weight.After 72 hours,rats with FBG≥16.7 mmol/L were considered to have successfully modeled T2 DM.Rats with successful modeling were divided into model group,Chinese medicine group and western medicine group.Rats in each group were intragastrically administered according to the corresponding dose.Rats in each group were sacrificed at the end of the 6th week of administration,and liver tissues were taken.The blood glucose,body weight,blood lipids and fasting insulin were detected.The morphological changes of liver tissues were observed by HE staining.The mRNA and protein expressions of PTP1 B,IRS-2 and PI3 K in liver tissues were determined by RT-PCR and Western blot.Results: 1.Compared with the blank group,the weight of the rats in the model group decreased rapidly,the amount of drinking water and the amount of urination increased significantly,the stool appeared soft and thin,the body hair gloss was weakened,and the symptoms of mental dysfunction and slow response were observed.After continuous drug intervention,compared with the model group,the above situation of the Chinese medicine group and the western medicine group were improved to varying degrees.2.Compared with the blank group,the blood glucose levels of the rats in each model group were significantly increased(P<0.01).After the 2nd,4th and 6th week of treatment,the FBG of the Chinese medicine group and the western medicine group were significantly decreased compared with the model group(P <0.01);There was no significant difference between Chinese medicine group and western medicine group(P>0.05).3.Compared with the blank group,the body weight of the rats in each model group decreased significantly(P<0.01).After the 2nd,4th and 6th week of treatment,the body weight of the Chinese medicine group and the western medicine group were significantly increased compared with the model group(P <0.01);There was no significant difference between Chinese medicine group and western medicine group(P>0.05).4.Compared with the blank group,the levels of TG and TC in the rats in each group were significantly increased(P<0.01).Compared with the model group,the TG and TC in the Chinese medicine group and the western medicine group were significantly lower.Statistical difference(P<0.01);there was no statistical difference between Chinese medicine group and western medicine group(P>0.05).5.Compared with the blank group,the FINS and HOMA-IR of the rats in each group were significantly increased,and there was significant difference between the two groups(P<0.01).Compared with the model group,the FINS and HOMA-IR of the Chinese medicine group and the western medicine group were significantly decreased.There was a statistical difference(P<0.01);there was no significant difference between the Chinese medicine group and the western medicine group(P>0.05).6.HE staining showed that the liver tissue of the model group showed fatty degeneration compared with the blank group.The fat cells of different sizes and sizes appeared in the serous cells of the liver cells,and even the necrosis showed fibrotic changes,while the liver tissue in the Chinese medicine treatment group improved to different degrees.7.Compared with the blank group,the mRNA and protein levels of PTP1 B in the liver tissues of each group were significantly increased(P<0.01),and the mRNA and protein levels of IRS-2 and PI3 K were significantly decreased(P<0.01).Compared with the model group,the Chinese medicine group and western medicine were compared with the model group.The mRNA and protein levels of PTP1 B in the liver tissue of the group decreased(P<0.01),and the mRNA and protein water of IRS-2 and PI3 K increased(P<0.01).There was no significant difference between the Chinese medicine group and the western medicine group(P> 0.05).Conclusion: 1.Traditional Chinese medicine compound Yitangkang can effectively reduce FBG in T2 DM rats and improve blood lipids.2.Traditional Chinese medicine compound Yitangkang has a certain repairing effect on the damaged liver.3.Traditional Chinese medicine compound Yitangkang acts as an inhibitor of PTP1 B and regulates the downstream proteins of its signaling pathway to improve IR,thus treating type 2 diabetes.
