Purpose:To observe the effect of traditional Chinese medicine compound Pidan Ning on the islet β-cell function and Bcl-2 expression in rats with impaired glucose regulation.Material and method:100 SPF Wistar male rats aged 60-70 days were selected and numbered according to their body weight.100 rats were divided into two groups by completely randomized block method: normal control group(20 rats)and impaired glucose regulation group(80 rats).The normal control group was fed with basic diet and impaired glucose regulation group was fed with high-sugar and high-fat diet.After 12 weeks,the rats with impaired glucose regulation were randomly divided into three groups: model control group(n=14),spleen Shengning treatment group(n=15)and metformin treatment group(n=14),and normal control group(n=14).After 4 weeks,the body weight,fasting blood glucose(FBG)and 2 h PG were measured before and after administration.The fasting insulin(FINS)level and the first phase pancreas were measured by high glucose clamp technique(HGCT).The insulin secretion level and the second phase insulin secretion level were used to evaluate the function of pancreatic beta cells.After the experiment,rats were killed and pancreatic tissues were taken from rats by laparotomy.The apoptosis of rat pancreatic islet cells was assessed by in situ end labeling(TUNEL).Bcl-2 in rat islet cells was detected by immunohistochemical staining.The expression of Bcl-2 was compared with the secretory function of rat islet beta cells,apoptosis of islet beta cells and the expression of Bcl-2.The correlation between the expression of Bcl-2 and apoptosis of islet beta cells was analyzed.Results:1.1.Islet beta cell secretion function: Before drug intervention,the insulin secretion levels of model control group,Pidan Ning treatment group and metformin treatment group were significantly lower than those of normal control group in the first and second phases(P <0.01),and there was no significant difference among model control group,Pidan Ning treatment group and metformin treatment group(P > 0.05);after drug intervention,the insulin secretion levels of Pidan Ning treatment group and metformin treatment group were significantly lower than those of normal control group(P < 0.01).The first phase of insulin secretion in metformin treatment group was significantly lower than that in normal control group(P < 0.01),higher than that in model control group(P < 0.05,P < 0.01),and the second phase of insulin secretion was also significantly lower than that in normal control group(P <0.01,P < 0.05),higher than that in model control group(P < 0.05,P < 0.01),and the second phase of metformin treatment group was significantly higher than that in Pidan Ning treatment group(P < 0.05).2.Apoptotic level of pancreatic islet cells: After drug intervention,the number of apoptotic islet cells in model control group,Pidan Ning treatment group and metformin treatment group was significantly higher than that in normal control group(P < 0.01),but the number of apoptotic islet cells in Pidan Ning treatment group and metformin treatment group was significantly higher than that in normal control group(P < 0.01).It was significantly lower than that of model control group(P < 0.01).3.Bcl-2 expression: The expression of Bcl-2 in Pidan Ning treatment group and metformin treatment group was significantly higher than that in model control group(P < 0.01),significantly lower than that in normal control group(P < 0.01),and there was no significant difference between Pidan Ning treatment group and metformin treatment group(P > 0.05).4.Correlation between Bcl-2 expression and islet cell apoptosis: bcl-2 protein level was negatively correlated with islet cell apoptosis(r=-0.847,P<0.01),ie,the higher the bcl-2protein level,The number of islet cell apoptosis is less.Conclusion:1.Pidan Ning can significantly improve the function of islet beta cells in rats with impaired glucose regulation.2.Pidan Ning can significantly inhibit apoptosis of pancreatic islet beta cells in impaired glucose regulation model rats.3.Pidan Ning can significantly increase the expression of Bcl-2 in blood of rats with impaired glucose regulation.4.Pidan Ning can improve islet β-cell function by increasing glucose-regulated Bcl-2expression in injured rat model,enhancing islet cell secretory function and reducing islet cell apoptosis. |
