| Objective: Doing electro-acupuncture at Zusanli(ST36)respectively on normal mice and TRPV1-/-mice for pre-stimulation,detecting lactate,creatine kinase and blood glucose level in mice,is to observe the preventive effect of electric acupuncture on the sport related fatigue of mice.Explore the mechanism of TRPV1 gene in sport related fatigue,and the effect on the action mechanism of acupuncture to prevent motor fatigue.Material and method:The mice are selected from 30 of Kunming wild mice and 18 mice with TRPV1 gene knockout.Group the mice using the random number table method.The Kunming wild mice are randomly divided into three groups of ten,setting as normal blank group,normal model group and normal acupuncture group;TRPV1 knockout mice are divided into three groups by this method,each group of six,setting as TRPV1 blank group,TRPV1 model group and TRPV1 acupuncture group.The blank group is fed normally without additional stimulation.After group completion,the normal acupuncture group and TRPV1 acupuncture group is given a seven-day electro-acupuncture intervention respectively,once a day,twenty minutes at a time,at 10:00--10:20,Using a 6805-A-type electric needle.Stimulus intensity 2-3ma,2-100hz(condensation wave-sparse wave),a total of seven times.Then,the model group and acupuncture group of two kinds of mice are simultaneously exhaustive swimming test.Confirm the molding success,blood sampling is performed for all 48 mice,then test the levels of lactic,creatinine and blood glucose in mice’s blood,statistical analysis of the results.Results: 1.Lactic acid content of normal mice Compared to normal mice model group and blank group serum lactic acid content,the lactic acid content increased significantly in model group,the difference is statistically significant(P<0.01).Comparing the lactic acid content of normal mice with the model group,lactic acid content decrease,it also has significant difference(P<0.01).2.CK content of normal mice The statistical analysis shows,the creatine kinase content in the normal mouse model group is higher than that in the blank group,the difference is statistically significant(P<0.01).Creatine kinase of normal acupuncture group content in mice have significantly lower than model group(P<0.01).3.Blood glucose levels The results show,the blood glucose of normal mouse model group is slightly lower than that of blank group,but there is no statistical significance(P>0.05).TRPV1 knockout mice are consistent with normal mice.,there is no significant difference between the blood glucose content and the blank group in the model group(P>0.05).4.TRPV1 gene knockout mice lactic acid content Statistical analysis of the TRPV1 gene knockout of mouse lactate,the serum lactate of these mice is significantly higher than that in the blank group(P<0.01);at the same time,contrast TRPV1 knockout mice acupuncture group and model group of serum lactic acid,lactic acid content decrease significantly,the difference is statistically significant(P<0.01).5.Comparison of lactic acid content between normal mice and TRPV1 gene knockout mice.Comparing to normal mice model with TRPV1 knockout mice model group serum lactic acid content,TRPV1 knockout mice have higher lactic acid level than normal mice,statistical analysis shows the difference is significant(P<0.01).Compare the TRPV1 knockout mice and normal mice acupuncture group of serum lactic acid level,the difference is statistically significant(P<0.05).6.TRPV1 gene knockout mice creatine kinase content TRPV1 gene knockout mouse model group showed a significant increase in creatine kinase content compared with the blank group(P<0.01).The serum creatine kinase content of TRPV1 gene knockout group is significantly lower than that in the model group.7.Comparing normal mice with TRPV1 gene knockout mice with creatine kinase content Statistical analysis of the experimental results shows,either the model group or the acupuncture group,there is no statistically significant difference in the content of creatine kinase between the two types of mice(P>0.05).Conclusion: 1.Electroacupuncture ST36 holes to prevent exercise-induced fatigue of exhaustion swimming mice.2.The TRPV1 inhibits the production of lactic acid in exhausted swimming mice.3.TRPV1 involves in the needle electroacupuncture ST36 of burnout prevention mechanism of lactic acid accumulation in mice.4.TRPV1 was not significantly associated with CK production in exhausted swimming mice.5.TRPV1 was not significantly associated with the prestimulation of electroacupuncture ST36 on the prevention of creatine kinase accumulation in exhausted swimming mice. |
