Purpose:Metastasis of tumor is the main cause of death of patients.Metastasis of tumor cells is a process involving many steps,including epithelial transformation,invasion of blood vessels,migration of blood vessels,exotion and tissue colonization,and epithelial transformation is a tumor cell.The precondition of the birth transfer.Therefore,finding a drμg to inhibit the occurrence of epithelial mesenchymal transition of tumor cells has become a hot topic in recent years.Traditional Chinese medicine for activating blood and removing stasis has achieved good curative effect in the treatment of malignant tumor with its effect of activating blood circulation,breaking blood stasis and eliminating the disease.The Chinese medicine,such as zedoary turmeric,three prism and cantharidis,can effectively inhibit the growth of tumor cells and be widely used in clinical.Elemene is an effective component extracted from Rhizoma curcumae,and it also has inhibitory effect on tumor cells.In this experiment,the inhibitory effect of elemene on lung cancer cell line H1299,breast cancer cell line MCF7 and colon cancer cell line SW480 was confirmed by studying the expression of elemene on the functional changes of tumor cells and the expression of protein level.Windows SPSS 22.0(IBM,Armonk,NY,USA)was used for all analyses,P < 0.05 was considered statistically significant.This experiment was funded by the Shenyang Science and Technology Plan Project(F16102400).Material and method:Human lung cancer cell line H1299,breast cancer cell line MCF7 and colon cancer cell line SW480 were all obtained from the Department of Pathology,China Medical University.Elemene injection was purchased from Dalian Jinang Pharmaceutical Co.,Ltd.(production batch number: 1612071).Lung cancer cell line H1299,colon cancer cell line SW480 and breast cancer cell line MCF7 were cultured in vitro,cultured in a 5% CO2,37°C incubator,exchanged for 1 to 2 days,and the cells grew to 80%to 90% confluent.At the same time,the concentration of 0,20,40,60 μg/mL elemene injection on H1299,MCF7 and SW480 cells,24 hours after the effect,the cloning assay wasused to detect the proliferation of cells in each group;transwell experiment to detect each group Cell migration and invasiveness;Western blot assay to detect epithelial-mesenchymal transition(EMT)related proteins including E-cadherin,N-cadherin,Snail and Vimentin,ZO1,Occludin and other protein expression levels.Results:1.With the increase of elemene injection concentration,the number of colony formation of three kinds of tumor cells decreased significantly,and the difference between elemene treatment group and blank group at different concentrations was statistically significant(P <0.05).The colony formation of H1299 cells in blank group(μg/ml)and drug treatment group(20 μg/ml,40 μg/ml,60 μg/ml)was 90±5,46±5,29±5,15±5,respectively;MCF7 cells were 135±5,78±5,60±5,40±5;SW480 cells were 150±10,130±10,100±10,and 82±10.2.With the increase of elemene injection concentration,the number of membrane cells in three tumor cells decreased significantly,and the migration and invasion of cells gradually weakened.Different concentrations of elemene treatment group compared with the blank group,the difference was statistically significant(P <0.05).The number of H1299 cells passing through the blank group(0 μg/ml)and drug treatment groups(20 μg/ml,40 μg/ml,60μg/ml)was 120±10,75±10,43±5,20±5,respectively;The number of membranes was 210±10,156±10,82±5,40±5,respectively;the number of SW480 cell membranes was 78±5,62±5,40±5,and 19±5.3.With the increase of elemene injection drug concentration,EMT-related protein expression:E-cadherin,ZO1,Occludin protein expression levels increased;N-cadherin,Snail,Vimentin protein expression levels decreased.Conclusion:1.elemene can inhibit the proliferation of cells,and its inhibitory effect increases with the increase of drug concentration.2.elemene can inhibit the migration and invasion of cells in a concentration-dependentmanner.3.elemene inhibits the migration and invasion of cancer cells by activating Snail expression,thereby increasing E-cadherin and down-regulating the process of N-cadherin.
