| Purpose:Observe the effect of Su zi Jiang qi soup on MUC5 AC and AQP5 in rat s with COPD and explore its mechanism from the perspective of airway mucus hyper secretion.Materials and Methods:1.Thirty-two SPF grade Wister male rats(body weight 250±20g)were randomly divid ed into blank group,model group,Fuluoshi group(abbreviated as Western medicine gr oup)and Suzi Jiangqi Decoction group(abbreviated as Chinese medicine group),each group 8,adaptive feeding for three days.2.Joint airway instillation of lipopolysaccharide smoked using methods established rat model of chronic obstructive pulmonary disease.The preparation method of the model:Each experimental group instilled 0.2 ml of lipopolysaccharide(1 mg/m L)into the ai rway of rats on the first and the 14 th day;on the 2nd day to the 13 th day,the 15 th day to the 30 th day.The rats were placed in a self-made smoke box to passively s moke,and the top and both sides of the smoke box were connected to the outside w orld with vent holes.Each time,8 rats were used 8 cigarettes and smoked for 30 mi nutes.Once a day,and evaluate the model on the 30 th day.After successful modeling,the blank group,model group,western medicine group,and traditional Chinese medic ine group were treated with gavage for two consecutive weeks.3.At the end of the gavage treatment,after observing and recording the general condi tion of the rats in each group,the rats were killed by anesthesia and left lung tissue was routinely taken for hematoxylin-eosin(HE)staining to observe the pathological ch anges of lung tissue in rats.The secretion of goblet cells in rat was observed by Alz heimer blue-periodic acid Schiff(AB-PAS)staining and the expression of MUC5 AC a nd AQP5 protein in rat lung tissue was observed by immunohistochemical staining.4.SPSS17.0 statistical software,the data were represented by "mean ± standard deviat ion"((?)± S),among groups were compared using ANOVA(one-way ANOVA),pairwi se comparisons using between groups T-test,P <0.05 was considered statistically signif icant.Results:1.COPD airway mucus hypersecretion model rats replicated successfully.2.The control group rats had a good mental state,normal activities,normal diet,stabl e breathing,shiny coat,normal coat color,and strong body.No obvious abnormal reac tion was observed.Rats in the model group and the blank group exhibited psychiatric exhaustion and decreased activity.As the smoking time prolonged,the rats developed cough,shortness of breath,weight loss,hair loss and loss of luster,and reduced feed ing and drinking.After treatment,the western medicine group and the Chinese medici ne group gradually improved their mental status,relieved the symptoms of cough and wheezing,gradually restored the luster of the hair,stabilized the breathing,and signifi cantly increased their activities.The food intake and body weight increased compared with before.3.Pathomorphological results showed that there was no thickening of the bronchial wa ll in the blank group,no goblet cell hyperplasia,no inflammatory cell infiltration unde r the submucosa,complete alveolar structure,and no ciliary detachment;bronchial epit helial cells in model rats Degeneration,necrosis,infiltration of inflammatory cells on t he wall of the tube,goblet cell hyperplasia,pulmonary bulla(bullous bullae)around t he lesion;symptoms of the model group rats after treatment with Fulu Shi and Suziji angqi Decoction improve,reduce the inflammatory infiltration,goblet cells decreased.4.Compared with the control group,MUC5 AC protein expression in model group incr eased,decreased level of expression of AQP5 protein,a significant difference(P <0.05);compared with model group,and the group administered Fu Lu Su lower gas In t he soup group,the expression of MUC5 AC was decreased and the expression of AQP5 was increased,and there was no significant difference between the two groups(P>0.05).Conclusion:1.Su zi Jiang qi soup can significantly reduce the symptoms of COPD model rats,a nd its mechanism may be related to anti-mucus hypersecretion.2.Fuluoshi and Suzijiangqi Decoction can all reduce the hypersecretion of airway muc ous in rats with chronic obstructive pulmonary disease.The mechanism may be achiev ed by down-regulating the expression of MUC5 AC protein and up-regulating the expre ssion of AQP5 protein. |
