| Purpose: To establish an AGA rat model by intra-articular injection of MSU and investigate the effect of topical plaster on the TLRs-MyD88 signaling pathway and IL-1β,TNF-α and inflammatory cytokines.To investigate the inhibitory effect of traditional Chinese medicine external ointment on the inflammatory response of AGA,and to elucidate the effect of traditional Chinese medicine external ointment on improving symptoms,and to provide scientific basis for clinical treatment of AGA.Material and method: 1.Forty male SD rats were randomly divided into five groups: normal group(A),model group(B),colchicine group(C),traditional Chinese medicine plaster group(D)Group of eight.Rats were housed in single cages,free-fed,and the experiment was started 1 week after feeding.The traditional Chinese medicine external ointment group and the futalin ointment group were applied externally with ointment 2 times/day and colchicine group 0.3 mg/kg/d.The other groups were given intragastric administration with saline for 5 days.On the third day before intragastric administration,the sodium uric acid crystals were injected into the joint cavity for injection.In addition to the blank group,all the other groups were modeled.The blank group was injected with normal saline in the joint cavity.2.Rats were anesthetized and sacrificed for 1 hour after the last gavage.Serum levels of IL-1β and TNF-α were measured by ELISA.The protein expressions of TLR2,TLR4 and MyD88 were detected by immunohistochemistry.HE staining was used to observe the pathological changes of ankle synovial tissue in rats.Results: 1.To observe the degree of swelling at 4 h,24 h,48 h,72 h after acute gouty arthritis induced by MSU in ankle joints of SD rats,which shows statistical difference(P <0.05)MSU induced acute gouty arthritis SD rat ankle swelling play a certain inhibitory effect.2.Herbal topical ointment can inhibit TLRs / MyD88 pathway upstream proteins TLR2,TLR4 and MyD88 in synovial tissue of ankle joint of acute gouty arthritis rats.Chinese medicine topical plaster can inhibit IL-1β and TNF-α in the serum of acute gouty arthritis rats.Conclusion: The curative effect of topical plaster for the treatment of acute gouty arthritis is curative effect,which may be related to the inhibition of TLRs-MyD88 pathway upstream proteins TLR2,TLR4,MyD88 and the downstream inflammation IL-1β,TNF-α. |
