Objective:Anxiety disorder is the most common psychiatric disorder.It is a neurological disorder characterized by anxiety.It is characterized by persistent anxiety,tension,panic and anxiety,accompanied by autonomic nervous disorder,muscle tension and motor disturbance.Anxiety has a serious impact on people’s quality of life.The side effects of common medication treatment are greater,and it also brings higher economic costs.Therefore,prevention and treatment of anxiety disorders from the perspective of daily diet is of great significance,which is also in line with the concept of "treating disease before disease" described in the Yellow Emperor’s Internal Classic.Tea is the most commonly used beverage in the world besides water.It has a long medicinal history.It has the functions of clearing the head,eliminating thirst,digesting,resolving phlegm,diuresis and detoxification.According to the fermentation degree,tea can be divided into green tea,black tea and oolong tea.In recent years,studies have found that green tea has anti-anxiety effect,but there are few reports about the anti-anxiety effect of black tea,oolong tea and other kinds of tea at home and abroad.Different kinds of tea contain different components,mainly in the different tea polyphenols,so it is speculated that the anti-anxiety effect is also different.Tea contains a variety of active ingredients.Its effect on mental diseases is the result of the combined action of caffeine,L-theanine and epigallocatechin gallate(EGCG).However,there have been many studies on L-theanine’s anti-anxiety effect,and less tea polyphenols.Epgallocatechin-3-gallate(EGCG)is a kind of polyphenols in green tea and oolong tea.It has strong antioxidant activity and can be used as food additive.In recent years,the neuroprotective effect,anti-Parkinson effect and anti-depressive effect of EGCG have been studied in the field of neuropharmacology,but there are few reports about its anti-anxiety effect and mechanism.Therefore,this topic mainly discusses the anti-anxiety effect and mechanism of green tea,oolong tea and black tea,and the anti-anxiety effect and mechanism of tea polyphenol monomer EGCG,in order to explain the anti-anxiety effect of tea,provide basis for the prevention and treatment of anxiety disorders from the perspective of daily diet,and provide reference for the development of EGCG.At the same time,this study also explored whether drinking tea when taking anti-anxiety Chinese medicine would affect its anti-anxiety efficacy,providing a reference for the rational use of tea and anti-anxiety Chinese medicine.Methods:1Comparison of anti-anxiety effects of green tea,oolong tea and black teaTo explore the effective dosage of tea for anti-anxiety,mice elevated plus-maze test,mice light/dark box test and mice open field test were used.The anti-anxiety effects of black tea(Yunnan Dianhong),green tea(Longjing West Lake)and oolong tea(Tieguanyin Anxi)were studied in mice behavioral test at the same dose.The contents of monoamine neurotransmitters NE,DA,5-HT and 5-HIAA in the brain of mice were detected by high performance liquid chromatography-electrochemical method,and the anti-anxiety mechanism of three kinds of tea was discussed and compared.2Anti-anxiety effect of EGCG and its mechanismThe anti-anxiety effects of EGCG were investigated by behavioral test in mice and empty bottle stimulation rats model.The levels of CRF,ACTH and CORT in serum of empty bottle stimulation rats were detected by ELISA,and the anti-anxiety mechanism of EGCG was explored from the perspective of HPA axis.3Effect of Oolong Tea on Anti-anxiety herb pair-Semen Ziziphi Spinosae-Fructus SchisandraeThe pharmacodynamics of Semen Ziziphi Spinosae-Fructus Schisandrae Chinensis herb pair(S W)and Oolong tea at the same time take and its single effect were compared by behavioral experiment in mice.Result:1Comparison of anti-anxiety effects of green tea,oolong tea and black tea1.1 Effective Antianxiety Dose of TeaThe water extract of green tea 600mg/kg(crude drug dosimeter)could significantly increase the percentage of open arm entries in EPM,the entries into the light box and the time spent in the light box in LDB,and the number of centrual entries and the time spent in centrual area in OFT.Therefore,the dosage of 600mg/kg was used to compare the anti-anxiety effects of the three kinds of tea.1.2 Comparison of Anti-anxiety Effects of Three TeasCompared with the blank group,oolong tea could significantly increase the percentage of open arm entries and the time spent in open arms in EPM,the entries into the light box and the time spent in the light box in LDB,and the number of centrual entries in OFT.Green tea could significantly increase the percentage of time spent in open arms in EPM,the time spent in the light box in LDB,and the number of centrual entries in OFT.Black tea could significantly increase the number of centrual entries in OFT.Among the three kinds of tea,the average value of each index of Oolong tea is the highest.1.3 Anti-anxiety mechanism of three kinds of teaCompared with the blank group,Oolong tea and green tea could significantly reduce NE content in the brain of mice,while black tea had a decreasing trend but no significant difference.Oolong tea could significantly reduce DA content in the brain of mice,while green tea and black tea had a decreasing trend but no significant difference,all three kinds of tea had a decreasing trend of 5-HT content in the brain of mice,but no significant difference.2 Anti-anxiety effect of EGCG and its mechanism2.1 Anti-anxiety effect of EGCG in miceCompared with the blank group,the medium dosage of EGCG(7.5mg/kg)and high dose(15mg/kg)significantly increased the percentage of time spent in open arms in EPM,and the low dose(3.75mg/kg)and medium dose(7.5mg/kg)significantly increased the percentage of open arm entries in EPM.Compared with the blank group,the medium and high dosage significantly increased the time spent in the light box in LDB,and the high dosage significantly increased the entries into the light box in LDB.Compared with the blank group,the medium dosage and high dose of EGCG can significantly increased the time spent in centrual area in OFT.2.2Anti-anxiety effect ofEGCG on Empty Bottle Stimulation ratsCompared with the model group,the middle dose(5.2mg/kg)and high dose(10.4mg/kg)of EGCG significantly increased the percentage of open arm entries in EPM of EBS rats,and the middle dose group significantly increased the percentage time spent in open arms.Compared with the model group,the middle dose group significantly increased the time spent in centrual area and number of centural entries in OFT of EBS rats.2.3Effect of EGCG on hypothalamus-pituitary-adrenal axis in Empty Bottle Stimulation ratsHigh dose of EGCG(10.4mg/kg)could significantly reduce the levels of CRF,ACTH and CORT in serum of rats.3Effect of Oolong Tea on Anti-anxiety herb pair-Semen Ziziphi Spinosae-Fructus SchisandraeCompared with the blank group,SW group and Oolong tea group significantly increased the percentage of time spent in open arms in EPM,among which SW group had the highest average value,while SWO group had a trend but no significant difference.Compared with the blank group,SW group and Oolong tea group can significantly increased the entries into the light box and the time spent in the light box in LDB,and the SWO group can significantly increased the entries into the light box in LDB,among which SW group had the highest average value.Compared with the blank group,SW group,Oolong tea group and SWO group all can significantly increase the number of centrual entries in OFT.Conclusion:At the same dosage,the anti-anxiety efficacy of Oolong Tea>Green Tea>Black Tea.The possible reasons are as follows;the influence on NE and DA systems in the brain by Oolong tea>green tea>black tea,while Oolong tea and green tea are rich in tea polyphenols,such as EGCG.EGCG has anti-anxiety effect,which may be related to the alleviation of hyperfunction of HPA axis.Tea may play an anti-anxiety role by affecting the contents of NE and DA in the brain,but not by affecting the 5-HT energy system.Drinking tea at the same time when taking anti-anxiety Chinese medicines such as Semen Ziziphi Spinosae and Fructus Schisandrae Chinensis may affect its efficacy. |
