| Objective:Serum endothelin(ET),nitric oxide(NO)and calcitin(CGRP)of migraine rats were investigated under the electro-acupuncture on acupoint of Shaoyang meridians(ES)in different periods.This study will provide some experimental basis for clinical application of acupuncture and moxibustion in the treatment of migraine.Methods:50 rats were randomly divided into five groups:(1)control,(2)model,(3)prevention of ES(PES),(4)treatment of SM(TES),(5)prevention and treatment of SM(PTES).The rats from PES and PTES group were bound and given the electro-acupuncture 30 min per day for 7 days.The other three groups were only bound at the same time.All rats were subcutaneously injected with nitroglycerin on day 8,while the control group was injected with the same volume of normal saline.After injection 30 min,the rats were bound 30 min.The TES and PTES group were simultaneously given electro-acupuncture.The behavior and 50% PWT were observed before and after modeling and treatment.The levels of ET and CGRP in serum were determined by enzyme-linked immunosorbent assay(ELISA).Furthermore,the level of NO in serum was examined by Nitrate Reductase.Results:1.At the end of modeling,the rats in each group showed redness in both ears,frequent forelimb scratches,increased number of cage climbing,vertical hair,tail biting and increased activity.The behavioral score increased significantly(P< 0.01),and the 50% PWT decreased significantly(P<0.01),indicating that the migraine model was successfully replicated.2.Behavioral comparison of rats in each group: After modeling,compared with group B,the behavioral score of group C and group E decreased(P<0.01).After treatment,compared with group B,the behavioral scores of group C,D and E were all lower(P<0.01);Compared with the control group(before treatment);the behavioral scores of C,D and E groups were significantly lower(P<0.01);there were significant differences between C,D and E groups(P<0.01),and the behavioral scores were C > D > E group.3.Comparison of 50% PWT in each group of rats: Compared with group B,50% PWT in group C and E increased(P<0.01).After treatment,compared with group B,50% PWT increased in groups C,D and E(P<0.01);Compared with the control group(before treatment),50% PWT in C,D and E groups increased significantly(P<0.01);there were significant differences between C,D and E groups(P<0.01),and 50% PWT in C < D < E group.4.Comparisons of ET,NO and CGRP contents in each group: Compared with group A,the contents of ET,NO and CGRP in group B increased(P<0.01).Compared with group B,the contents of ET,NO and CGRP decreased in groups C,D and E(P< 0.01).Compared with group C,the contents of ET,NO and CGRP in group E decreased(P<0.01),but there was no significant difference in the contents of ET,NO and CGRP in group D(P>0.05).Compared with group D,the contents of ET,NO and CGRP in group E decreased(P<0.01).Conclusion:1.Electro-acupuncture at Shaoyang meridian points can relieve the pain of migraine rats,decrease the expression of ET,NO,CGRP and other vasoactive substances to increase pain threshold,which may be one of the mechanisms.2.The method of prevention and treatment of electro-acupuncture at Shaoyang meridian points is better than that of prevention and treatment at acute stage,suggesting that the method of prevention and treatment of electro-acupuncture at Shaoyang meridian points can be used as an advantageous scheme,providing certain experimental basis for clinical treatment of migraine. |
