Objective:Suspension tail method is adopted to establish the male rat tail suspension disuse osteoporosis model,modeling after successful give rhizoma drynariae lavage experimental intervent ion,total flavonoids in alendronate sodium phosphate lavage for positive control,in a standard laboratory animal drinking water to fill the stomach as the negative control group and with free activity as blank control group,experimental group and the control group of rats bone mineral density and histomorphology observation and tibia bone metabolism growth signaling pathways important Runx-2 mrna expression of transcription factors,trying to evaluate rhizoma drynariae total flavonoids of tail suspensi on waste use sex male osteoporosis prevention and treatment in rats,to explore its possible mechanism of action of targets,To further expand the study on the effect of total flavonoids from gusuan bubao on the model of osteoporosis in male rats with disused tail suspension,so as to provide more theoretical basis for the effective treatment of disused osteoporosis in clinical practice.Methods:All 48 6-month old SPF male experimental rats were randomly divided into 4 groups after 1 week of adaptive feedin g,namely,free exercise group(blank control group),tail-hanging drinking water group(negative control group),tail-hanging alendronate sodium group(positive control group),tail-hanging bone bone supplementation total flavonoids group(experimental group),12 rats in each group.The tail suspension drinking water group,the tail suspension arondronate sodium group and the tail suspension bone crushing total flavonoids group were used for modeling,and the blank control group was not treated.For each o f the three groups,total flavonoids,sodium aron phosphate and standard drinking water of experimental animals were given to the rats by gavage for 28 days.28 days after the tail suspension abdominal cavity by chloral hydrate anesthesia,then take the tibia and the femur proximal specimens,in vitro proximal femoral bone mineral density measurement,observe the tibia organization form,and USES the method of real-timePCR Runx in the femoral group carries on the quantitative analysis and comparison to 2,explore Runx-2 in tail suspension under the action of total flavonoids in rhizoma drynariae disuse osteoporosis in the male rat model of mRNA expression level.Results:1.Compared with the negative control group,the bone trabeculae in the total flavonoids group were more evenly distributed,orderly and continuous,with a certain degree of benign performance in terms of quantity,distribution range,quantity and structure thickness.2.Compared with the blank control group,bone mineral density an d Runx-2 mRNA expression levels in the proximal femur of each group were decreased,with statistically significant differences(P < 0.05).3.Compared with the negative control group,bone mineral density and Runx-2 mRNA expression were increased in the pr oximal femur of the two groups.The difference was statistically significant(P < 0.05).4.There was no significant difference in proximal femoral bone density and Runx-2 mRNA expression between the two groups(P > 0.05).Conclusion:The expression of Runx-2 mRNA,an important transcription factor of bone metabolic growth signaling pathway,was up-regulated in the model of disused tail bone osteoporosis in male rats,and bone mineral density was increased.It can also improve the tibial histomorphology,which is helpful to promote the treatment of disused osteoporosis with total flavonoids.
